Abstract
Omurilik, beyin ve vücudunuzun kalanı arasında sinir iletimini sağlayan bağlantı yoludur. Omurilik hasarları genellikle spor veya araba kazaları gibi ani travmalar sonucunda, omurganın kayarak veya parçalanarak omurilik dokusunun içine girmesi ve nöral hücrelere zarar vermesi nedeniyle elektrokimyasal dolaşımın kesilmesine sebep olur. Bu durum, yaralanmanın olduğu yerden aşağıda kalan tüm vücutta duyu ve motor kontrolünün kaybolmasına yol açar. Travmanın ikincil etkisine bağlı olan kan ihtiyacındaki değişiklik, bağışıklık yanıtı, serbest radikallerin ve uyarıcı nörotransmitterlerin artması gibi sebeplerden dolayı, omurilik yaralanmasının olduğu yer ve çevresinde birçok hücre ölür. Merkezi sinir sisteminde nörojenez var olmasına rağmen aşırı hücre ölümü ve kronik safhada ortaya çıkan glial yara nedeniyle endojen rejenerasyon yetersizdir. Omuriliği hasar almış kişiye uygulanan mevcut tedavi yöntemleri fonksiyonel onarımdan ziyade komplikasyonları önlemeye yöneliktir. Hasarın birincil ve ikincil fazında uygulanan ilaç tedavileri sonrası hasta aylar ya da yıllar süren fizik tedavi ve rehabilitasyon sürecine tabii olmaktadır. Psikolojik ve ekonomik açıdan ağır olan bu hastalıkta iyileşme oranı % 1'in altındadır. Doku mühendisliği bu hastalığın tedavisinde en umut verici alandır. Uygun bir biyomalzeme doğal omuriliğin yapısına benzer özellikler göstererek nörojenez için uygun mikroçevre sağlayabilir. Grafen üstün özelliklere sahip karbon allotroplarından biridir. Çok iyi elektrik iletkenliği ve mekanik dayanım göstermektedir. Sinir sistemi elektriksel faaliyetlere dayanan fonksiyonlar gösterdiğinden grafen nöral modeller için ideal bir malzemedir.Bu tez çalışmasında grafen bazlı doku iskelesi hazırlandı ve karakterize edildi. Grafitten kimyasal eksfoliasyonla elde edilen grafen oksit freeze-drying yöntemi ile üç boyutlu yapıda doku iskelesi elde edilmesinde kullanıldı. Farklı karbon/oksijen oranına sahip grafen oksit elde edilmesi amacıyla termal ve kimyasal indirgeme gerçekleştirildi. Vakum filtrasyon tekniği ile hazırlanan film yapılı (iki boyutlu) grafen oksitlerin elektriksel iletkenliği ölçüldü. Sentezlenen grafen oksit FTIR, AFM, UV spektrofotometre ve SEM-EDS ile karakterize edildi. Doku iskelesinin SEM görüntüleri üç boyutlu ve poroz yapıda doku iskelesi elde edildiğini göstermektedir. Termal ve kimyasal indirgeme ile elde edilen grafen oksitlerin C/O oranı sırasıyla 9,52 ve 27,93; iletkenlik değerleri ise 190,5 ve 2831,65 S/m olarak belirlendi. Elektrik iletkenlik analiz sonucu grafen oksitin omurilik dokusuna benzer iletkenlik değerine sahip olduğunu göstermiştir. Spinal cord is the connection that provides the nerve conduction between the brain and the rest of the body. Spinal cord injuries usually cause the interruption of electrochemical circulation due to the vertebrae to slip or break into the spinal cord and damage the neural cells by sudden traumas such as sports or motor-vehicle accidents. This leads to the loss of sensory and motor control in the entire body below the injury site. Because of the secondary effects of trauma such as changes in blood requirements, immune response, increased free radicals and stimulating neurotransmitters, many cells die in the area and around the spinal cord injury. Although central nervous system has neurogenesis, endogeneous regeneration is unsufficient due to the massive cell death and emerging glial scar in cronic phase. Current treatment methods for spinal cord injured individuals are aimed at preventing complications rather than functional repair. After the drug treatments applied in the primary and secondary phase of the injury, the patient is subjected to physical therapy and rehabilitation for months or years. In this psychologically and economically severe disease, the cure rate is below 1%. Tissue engineering is the most promising field in the treatment of this disease. A suitable biomaterial can provide the appropriate microenvironment for neurogenesis by displaying properties similar to that of the native spinal cord. Graphene is one of the carbon allotropes with superior properties. It has very good electrical conductivity and mechanical strength. As the nervous system shows functions based on electrical activities, graphene is the ideal material for neural models. In this thesis study, a graphene-based tissue scaffold was prepared and characterized. The graphene oxide obtained by chemical exfoliation from graphite, which was used to obtain scaffold with three dimensional structure by freeze-drying method. Reducing process by thermal and chemical was made to obtain graphene oxide with different carbon/oxygen ratio. The electrical conductivity of the film-structured (two-dimensional) graphene oxide prepared by the vacuum filtration technique was measured. Synthesized graphene oxide was characterized by FTIR, AFM, UV spectrophotometerand SEM-EDS. SEM images of the scaffold showed that the scaffold with three-dimensional and porous structure was obtained. C/O ratio of thermal and chemical reduced graphene oxide was determined 9,52 and 27,93 respectively; and electrical conductivity of it was determined 190,5 and 2831,65 S/m, respectively. Electrical conductivity analysis showed that graphene had the similar conductivity value as the spinal cord tissue.
