Abstract
VI ÖZET Glikoz izomeraz gıda endüstrisinde glikozu fruktoza çevirmekte yaygın olarak kullanılan bir enzimdir. İzomerizasyon reaksiyonu ile yüksek sıcaklıklarda daha çok fruktoz üretilebildiği için endüstrinin termostabil glikoz izomerazlara olan ilgisi gittikçe artmaktadır. Bu çalışmada bakteri tararından hücre dışına salgılanabilen termostabil glikoz izomerazın üretilebilmesi için gerekli rekombinant DNA teknikleri araştırılmıştır. Uygulanan stratejide, glikoz izomeraz geni üretilen proteinin hücre dışına salgılanmasını sağlayan bir gen dizisi içeren füzyon vektörüne eklenmiştir. Glikoz izomeraz geni Thermus (hemophilus (ATCC 27634) hücrelerinin DNAsından PCR yöntemiyle çoğaltılmış ve pEZZ18 füzyon/salgılama vektörünün Psil kesim bölgesine eklenmiştir. Rekombinant vektör Escherichia coli HB101, TBI, XL-1 ve DH1 suşlanna transforme edildiğinde hücre büyümesinin önemli ölçüde azaldığı, hatta bazı suşların büyümesinin tamamen durduğu gözlemlenmiştir. Rekombinant plazmidi içeren hücrelerden çeşitli yöntemlerle elde edilen proteinlerin SDS-PAGE analizi yapıldığında, beklenen büyüklükte gözlemlenen belirgin bir protein bandı füzyon proteininin ancak çok az bir miktarının periplazmik aralığa taşınabildiğim proteinin daha çok sitoplazmada kaldığını göstermektedir. ABSTRACT Glucose isomerase is an enzyme widely used in the food industry to catalyze the conversion of glucose to fructose. The isomerization reaction is reversible and a higher temperature gives a higher fructose content. Hence there is a continuing interest in thermostable glucose isomerase enzymes. In the present study genetic engineering techniques are exploited to produce a thermostable glucose isomerase as a secretion product from bacteria. The strategy adopted was to attach the glucose isomerase gene to a fusion vector with a leader sequence for secretion into the culture medium. The gene for glucose isomerase from Thermus thermophilic (ATCC 27634) was amplified using PCR and inserted into the unique PstI site of the fusion/secretion vector pEZZ18. It was observed that the cell growth was highly impaired when the recombinant vector was transformed into Escherichia coli strains HB 101, TBI, XL-1 and DHL Eventually, the viability of the cells was abolished. SDS- PAGE analysis of the proteins extracted from the whole cells harbouring the recombinant plasmid exhibited a band in the expected size of the fusion protein, indicating that the fusion protein mainly remained in the cytoplasm and very little was transported to the periplasmic space.
