ÖZETTezin cinsi : DoktoraTezin adı : Leucojum aestivum L.' da sitolojik ve sitoembriyolojik çalışmalarÜniversite : Trakya ÜniversitesiEnstitü : Fen BilimleriAnabilim dalı : BiyolojiProgramı : Hücre BiyolojisiBu araştırmada Edirne-Tavuk Ormanı'nda doğal olarak yetişen Leucojum aestivum L.sitolojik ve sitoembriyolojik yönden ışık, floresans ve transmisyon elektron mikroskopiyöntemleri kullanılarak incelenmiştir. L. aestivum'un kromozom sayısı 2n=22'dir. Anterlertetrasporangiattır. Anter çeperi epidermis, endotesyum, ara tabaka ile salgı tapetumundanoluşur. Tapetum hücrelerinde, normal mitoz bölünme, sekonder nukleus bölünmeleri ve nukleusbirleşmeleri saptanmıştır. Mikrosporogenez ve polen mitozu genellikle düzenlidir. Olgun polentaneleri 2 hücrelidir. Polen sterilitesi %1.7 ve polen tanelerinin in vitro ortamdaki çimlenmeoranı %73'tür. Olgun anter çeper hücrelerinin polisakkarit içeriği daha zengindir. RNA içeriğiise anter çeper hücreleri ve mikrosporların gelişim evrelerine göre farklılık göstermektedir.TEM ile yapılan incelemelerde mikrosporogenezin ve polen mitozunun erken evrelerindePAH'inde ve mikrosporlarda organel içeriği yönünden polar bir dağılım görülmüştür. Profaz Ievresinde PAH'leri arasında sitoplazmik kanallar gözlenmiştir. Tetrad evresinde tapetumda aktifsekresyon yapılır ve tapetum dejenere olmaya başlar. L. aestivum'un ovulleri anatrop, bitegmikve krassinusellat tiptedir. Mikropil iç integümentten oluşur. Arkespor hücresi doğrudanmegaspor ana hücresini verir. Embriyo kesesi gelişimi bisporik Allium tipindedir. Sinergidhücrelerinde filiform aparatus bulunur. Polar nukleuslar fertilizasyondan önce birleşereksekonder nukleusu antipodlara yakın bölgede oluştururlar. L. aestivum' da megasporogenezevresindeki megasporangiyumda, polisakkaritler kalaza ve nusellus dokusu hücrelerinde,megagametogenez ve olgun megagametofit evresinde ise dış integüment ile kalaza hücrelerindegörülmüştür. Its chromosome number is 2n=22. Anthers aretetrasporangiate. Anther wall formed with an epidermis, endothecium, middle layer andglandular tapetum. Normal mitosis, secondary nucleus divisions and fusions were determined intapetum cells. Microsporogenesis and pollen mitosis are generally regular. Mature pollen grainsare 2-celled. Pollen sterility is 1.7% and germination ratio of pollen grains in vitro is 73%.Mature anther wall cells? polysaccharide content is more than immature ones. RNA contentshows difference according to anther wall cells and microspores? developmental stages. Duringearly stages of microsporogenesis and pollen mitosis, organelle contents of microspore mothercells (MMC) and microspores were shown polarity when they examined in TEM. Cytoplasmicchannels were observed between MMC in prophase I. Active secretion is done in tapetum andtapetum begins to degenerate in tetrad stage. L. aestivum?s ovules are anatropous, bitegmic andcrassinucellate type. Inner integument forms micropyle. Archesporial cell developed directlyinto a megasporocyte. Embryo sac development is bisporic Allium type. Filiform apparatus isobserved in synergids. Polar nuclei fuse before fertilization to form secondary nucleus near theantipodals. Polysaccharides have been seen in chalazal and nucellar cells of megasporangium atmegasporogenesis and in outer integument and chalazal cells at megagametogenesis and maturemegagametophyte stages.
