Huş afidi (Euceraphis punctipennis Zetterstedt)`nin biyolojik mücadelesinde Lecanicillium muscarium (Petch) zare and gams`un etkinliği ve fungusun farklı sıcaklıklarda gelişimi

Abstract
Bu çalışma, Erzurum ilinde park ve bahçelerde bulunan huş ağaçlarındaki en önemli zararlılardan biri olan Euceraphis punctipennis (Zetterstedt) (Hemiptera: Aphididae)'e karşı, entomopatojen fungus Lecanicillium muscarium'un etkinliğinin belirlenmesi amacıyla yapılmıştır. Huş ağaçlarında afit bulunan yapraklar alınarak laboratuara getirilip, üzerinde en az 10'ar birey bulunan yapraklar petri kaplarına konulmuştur ve üzerine değişik konsantrasyonlarda (1x106, 1x107 ve 1x108 konidi/ml) fungus sporları sprey edilmiştir. Afit ölümleri 1. gün başlamış 1x106 konidi/ml'de 5. gün, 1x107 konidi/ml'de 6. gün, 1x108 konidi/ml'de 4. gün %100'e ulaşmıştır. Fungusun etkinliğinin doğal koşullarda da belirlenmesi için en etkin konsantrasyon olan 1x107 konidi/ml kullanılmıştır. Konukçu bitkinin dallarında, en az 10'ar afit bulunan yapraklara hazırlanan süspansiyon sprey edilmiş ve bu yapraklar tülbent kafeslere alınmıştır. Bu uygulama 10 tekerrürlü olarak yapılmıştır. Hem laboratuar hem de arazi şartlarında kontrol amacıyla sdH2O sprey edilmiştir. Ölen afit sayımı günlük olarak yapılmış, ortalama ölüm süreleri (LT50) belirlenmiştir. Ölümler 1. gün başlamış ve 5. gün %100'e ulaşmıştır. Hem laboratuar hem de doğal koşullarda ölen bireylerden fungus reizolasyonları yapılmıştır. Bu bulgular L. muscarium'un E. punctipennis'in mücadelesinde kullanılabileceğini göstermektedir.Diğer taraftan, fungusun değişik sıcaklıklardaki (20, 25, 30, 35, 40°C) gelişme hızı incelenmiştir. En iyi fungus gelişiminin 25°C'de olduğu, 35 ve 40°C sıcaklıklarda herhangi bir fungus gelişmesi olmadığı gözlenmiştir. This study was conducted to determine the effectiveness of an entomopathogen fungus, Lecanicillium muscarium, against Euceraphis punctipennis (Zetterstedt) (Hemiptera: Aphididae) which is one of the most important pests on birch trees in the parks and gardens in Erzurum Province. The leaves having aphids collected from birch trees were brought to laboratory, put into petri dishes with 10 individuals at least, and the fungus spores in various concentrations (1x106, 1x107 ve 1x108 conidia/ml) were sprayed on the leaves. Aphid deaths began at the first day, and increased to 100% on the fifth, sixth and fourth days, in the concentrations of 1x106, 1x107 and 1x108, respectively. The most effective concentration, 1x107, was used to determine the effectiveness of fungi under the natural conditions. The suspension prepared was sprayed on the leaves with 10 aphids at least on the host plant branches, and taken into net cages. This application was made with 10 replications. For the control, sterile deionized water (sdH2O) was sprayed under both laboratory and field conditions. Counting of dead aphids were made by daily and letal time (LT50) was determined. Deaths began on the first day and increased to 100% on fifth day. Fungus reisolations from the death individuals under the laboratory and natural conditions were made. These results showed that L. muscarium will be able to use in the control of E. punctipennis.On the other hand, growth rate of the fungus at different temperatures (20, 25, 30, 35, 40°C) was investigated. It was observed that the best fungal growth was at 25°C, and there was no fungal growth at 35 and 40°C.
