Abstract
?Radyoprotektör? olarak adlandırılan bazı kimyasal maddelerin organizmayı iyonizanradyasyonun zararlı etkilerinden koruyabildigi bulunmustur. Bu maddeler çok küçükmiktarlarda bile organizmayı iyonizan radyasyonun öldürücü etkilerinden koruyabilirler.Günümüze kadar çok çesitli maddelerin radyoprotektif etkileri üzerine birçok arastırmayapılmıs ancak polimerik yapıda bir radyoprotektöre rastlanmamıstır. Radyoprotektif etkisiarastırılan bu maddeler küçük molekül agırlıkları nedeniyle organizmada kısa süreli biyolojiketki göstermektedirler. Bu nedenle polimerik radyoprotektörlerin gelistirilmesi çok büyükönem tasımaktadır.Bu amaçla, suda çözünen negatif yüklü poliakrilik asit (PAA) ve akrilik asit vinilpirolidonkopolimerleri (P(AA/VP), nötral polivinil pirolidon (PVP) ve bunların Cu(II) ile iyonkoordinasyon bilesenlerinin ve kan proteinleri içersinde en çok miktarda bulunan proteinfraksiyonu Serum Albumini (BSA) ile olusturdukları çesitli kompleksler kullanıldı.Radyasyon etkisi ile komplekslerde meydana gelen degisimler kimyasal yöntemlerle (uv-vis,FT-IR spektrofotometri, HPLC, elementel analiz, AAS) incelendi. Bu maddelerin radyasyonvarlıgında, dolasan kan lenfositleri üzerinde olusturdukları etkiler ise sitogenetik yöntemler(mikronükleus analizi (MN), kromozom aberasyon (CA) analizi) kullanılarak arastırılmıstır.Biyolojik etki mekanizmasının anlasılabilmesi için polimerlerin farklı molekül agırlıklarınınve konformasyonunun denenmesi ile ilgili çalısmalara agırlık verilmistir.Belirli bir radyasyon dozuna kadar polimer-metal ikili kompleks sisteminde Cu2+'ninradyasyon etkisi ile Cu1+'e indirgenmesi, artan radyasyon dozu ile artıs göstermistir. Busonuçlar radyasyon enerjisinin ikili kompleksdeki Cu2+ iyonları tarafından absorblandıgıdüsüncesini dogrulamıstır. Sitogenetik çalısmalardan ise molekül agırlıgı ve konformasyondegisiminin hücreye olan etkisinin farklı oldugu gösterilmistir. PAA ve komplekslerinin hücreile olan etkilesiminde interpolimer kompleks olusumu ve kooperatif etkilesimin rolü oldugugösterilmistir. PAA'nın belirli bir konsantrasyon aralıgında radyoprotektif bir etki gösterdigibelirlenmistir.Bu çalısmalar mevcut küçük molekül agırlıklı, organik radyoprotektif maddelere alternatifolabilecegi düsünülen daha etkili, düsük toksisiteye sahip radyoprotektörlerin gelistirilmesinebir baslangıç olması amacı ile yapılmıstır.Anahtar Kelimeler: Radyoprotektör, Suda çözünen polimer, PAA, polimer-metal kompleksi,MN, CA. It has been found that some chemical substances, which is taken in organism before exposureto ionizing radiation, protect the organism against lethal and harmful effect of ionizingradiation called as ?radioprotector?. They protect the organisms against a lethal dose ofionizing radiations when they are also extremely small amount. Various compunds have beeninvestigated towards on radioprotective effect but polymer containing have not beendiscovered up to now. Since the organic compounds investigated radioprotective effect haverelatively less molecular weight, so that they show biological effect in a short time. That iswhy it is very important to improve polymeric radioprotector.In the present work, ion-coordination compounds of some water soluble negatively chargedpolyacrylic acid (PAA) and copolymer of acrylic acid vinylpyrrolidone (P(AA/VP), neutralpolyvinylpyrrolidone (PVP) mediated with Cu(II) and Bovine Serum Albumin (BSA) whichis found the highest protein fraction in blood have been used. Chemical methods (uv-vis,FT-IR spectrophotometry, HPLC, elemental analysis, AAS) have been employed to thesecomplexes in order to investigate the effect of irradiation. The effect of radiation on in vitrohuman peripheral lymphocyte in the presence of these compouds have been researched usingcytogenetic methods (micronucleus analysis (MN), chromosome aberration (CA) analysis).My work has been focused on polymers having several different molecular weights andconformations in order to understand the biological effect mechanism.Until certain radiation doses, reduction of Cu(II) ion to Cu(I)ion in polymer-metal complexsystem by rising radiation effect has increased. These results have confirmed, the radiationenergy was absorbed by Cu(II) ion in polymer-metal complex system. It has been showed thatin cytogenetic studies, the effect of molecular and conformational alterations were differenceon cell. The role of interpolymer complex formation and cooperative interaction have beenpointed out in the interaction of PAA and their complexes with cell. It has been determinedthat, in specific PAA concentration has radioprotective effect.The objective of this study is to develop high-effective polymer containing radioprotectorswith low toxicity as an alternative to the conventional radioprotectors mentioned before.Keywords: Radioprotector, water soluble polymer, PAA, polymer-metal complex, MN, CA.
