Abstract
Bu çalışmada, toksik olduğu bilinen kadmiyumun eser seviyelerde ekonomik ve basit bir yöntem ile tayin edilebileceği yüksek doğruluk ve duyarlılık bir analitik yöntem geliştirilmesi amaçlanmıştır. Bu kapsamda, öncelikle Dağıtıcı Sıvı-Sıvı Mikroekstraksiyon (DSSME) tekniği uygulanarak gerçek numunelerdeki kadmiyum önderiştirilmiş ve sonrasında Yarıklı Kuvars Tüp (YKT) entegre edilmiş Alevli Atomik Absorpsiyon Spektrofotometresi (AAAS) sisteminde yüksek duyarlılıkta tayin edilmiştir. Analitik yöntem geliştirilirken DSSME tekniğinin ve YKT-AAAS sisteminin değişken tüm parametreleri optimize edilmiştir. DSSME tekniği, numunedeki kadmiyum iyonlarının difenilkarbazon (DFK) ile kompleks oluşturması, oluşan komplekslerin numunede çözünmeyen bir organik faz yardımı ile ayrılması ve kompleksin parçalanarak serbest kadmiyum iyonlarının tayin edilmesi adımlarını içermektedir. DSSME yönteminde önderiştirme veriminin yükseltilmesi amacıyla bu adımlardaki değişken parametreler olan DFK konsantrasyonu, ekstraksiyon öncesi karıştırma süresi, tampon çözeltinin pH'sı/miktarı, dağıtıcı çözücü türü/miktarı, ekstraksiyon çözücüsü türü/miktarı, tuz türü/miktarı ve karıştırma türü/süresi optimize edilmiştir. Bununla birlikte YKT-AAAS sisteminde kadmiyuma ait sinyali yükseltmek amacıyla sistemi etkileyen örnek akış hızı, alev hızı ve YKT yüksekliği parametreleri de optimize edilmiştir.Geliştirilen analitik yöntemin performansı doğrusallık, gözlenebilme limiti (GL), tayin limiti (TL), yüzde geri kazanım (%GK) ve doğruluk kriterleri bakımından incelenmiştir. Buna göre DSSME-YKT-AAAS birleştirilmiş yöntemin gözlenebilme ve tayin limitleri sırarıyla 0.45 ve 1.46 ng/mL olarak tespit edilmiştir. Geliştirilen yöntem 0.999 korelasyon katsayısı (R2) ile 1.0 - 17.5 µg/L arasında iyi bir doğrusallık göstermiştir. Klasik AAAS sistemine kıyasla, YKT-AAAS, DSSME-AAAS ve DSSME-YKT-AAAS sistemleri için tayin gücünde sırasıyla 3.5, 31.6 ve 95.5 kat artış sağlanmıştır. Doğrusal aralıktaki en düşük konsantrasyon değerinin 6 tekrarlı ölçümleri ile edilen düşük yüzde bağıl standart sapma (%BSS) değeri yöntemin tekrarlanabilir olduğunu göstermiştir. Geliştirilen analitik yöntemin doğruluğu atık su sertifikalı referans maddes (EU-L-2) kullanılarak test edilmiş ve elde edilen sonuçların sertifikalı değerle uyum içerisinde olduğu tespit edilmiştir bu da yöntemin doğruluğunu kanıtlamıştır. Ayrıca yöntemin bazı gerçek örneklere uygulanabilirliğini test etmek amacıyla İstanbul'daki üç farklı ilçeden alınan musluk suları ve belediye atık suyunda geri kazanım çalışmaları yürütülmüştür. Geri kazanım sonuçlarının % 100'e yakın olması, analitin yüksek doğruluk ve duyarlılıkta tayininde matriks kaynaklı bir giriş etkisinin bulunmadığını göstermiştir. Anahtar Kelimeler: Kadmiyum, mikroekstraksiyon, DSSME, YKT, AAAS In this study, an economical and simple analytical method was aimed to be develop for determination of the toxic element cadmium at trace levels with high accuracy and sensitivity. In this context, Dispersive Liquid-Liquid Microextraction (DLLME) was used to preconcentrate cadmium from real samples and determined at high sensitivity with a Slotted Quartz Tube (SQT) integrated Flame Atomic Absorption Spectrophotometer (FAAS) system. In the development of the analytical method, all variable parameters of the DLLME method and the SQT-FAAS system were optimized. The DLLME method involved complexation of cadmium ions in the sample with diphenylcarbazone (DPC), separation of the resulting complex in the sample with the aid of an insoluble organic solvent, and cleavage of the complex to determine free cadmium ions. In order to increase the preconcentration efficiency in the DLLME method, DPC concentration, mixing period before extraction, pH/volume of the buffer solution, dispersive solvent type/amount, extraction solvent type/amount, salt type/amount and the mixing type/period were all optimized. In addition to these, the sample flow rate, fuel flow rate and SQT height which affect the SQT-FAAS system were also optimized in order to increase the signal of cadmium.The performance of the developed analytical method was examined in terms of linearity, limit of detection (LOD), limit of quantitation (LOQ), percent recovery and accuracy criteria. Accordingly, the limits of detection and quantitation of the DLLME-SQT-FAAS combined method were found to be 0.45 and 1.46 ng/mL, respectively. The developed method showed good linearity between 1.0 and 17.5 µg/L with a 0.9991 correlation coefficient (R2). Compared to the conventional FAAS system, the SQT-FAAS, DLLME-FAAS and DLLME-SQT-FAAS systems recorded 3.5, 32 and 96 times increase in detection power, respectively. The low percent standard deviation (%RSD) value obtained from 6 replicate measurements of the lowest concentration in the linear calibration range confirmed the methods precision. The developed method was applied to the waste water certified reference material (EU-L-2), and the closeness of experimented results to the certified value validated the method's accuracy. In addition, recovery studies were carried out in order to determine the applicability of the method to some real examples, tap water from three different provinces in Istanbul and municipal waste water. The recovery results were close to 100%, indicating that there was no matrix-based effect in the determination of the analyte with high accuracy and sensitivity.Keywords: Cadmium, microextraction, DSSME, SQT, FAAS
