Berberis vulgaris L. üzerinde HPLC ile analitik çalışmalar

- 53 - VI ÖZET Berberis vulgaris L. bitkisinin kurutulmuş ve toz edilmiş köklerinin alkollü ekstresinden hareketle hazırlanan kloroformlu çözeltiler kullanılarak HPLC ve UV-VIS spektroskopisi yöntemleriyle alkaloit analizleri yapıldı. Silika kolon (Alltech Econosphere Silika 5U Column) ve hareketli faz olarak 0.005 M sodyum asetat içeren metanol: 1-4 dioksan : asetik asit (8.8:1:0.2) kullanılarak yapılan HPLC analizinde UV dedektör 385 nm dalga boyuna ayarlandı. Drogdakİ ana alkaloit olan Berberin miktarı %2.4 olarak saptandı. Benzer şekilde hazırlanan kloroformlu çözeltilerin 385 nm dalga boyunda yapılan UV spektroskopik analizinde drogdaki alkaloit miktarı % 3.1 olarak belirlendi.

- 54 - VII SUMMARY Alkaloid analysis of Berberis vulgaris L. have been carried out by HPLC and UV-VIS spectroscopic methods in the chloroform solutions which were prepared from alcoholic extracts of powdered dry roots of the plant. The working wavelength of UV detector of HPLC has been adjusted to 385 nm and 0.005 M sodium acetate in methanol 1-4 dioxane : acetic acid (8.8:1:0.2) mixture has been used as the mobil phase, the column packing material was silica (Alltech Econosphere Silica 5U Column). It has been determined that Berberin, the major alkaloid in the drog was 2.4 %. In UV spectral analyses carried out at 385 nm, alkaloid content of the drug has been found 3.1 % in the chloroform solutions which were prepared in the same manner.
