Abstract
93 ÖZET Bu çalışmada hücre migrasyonu, proliferasyonu ve diferansiyasyonunda önemli etkileri olduğu saptanmış olan hyaluronik asitin diş pulpası üzerine kapaklama ajanı olarak kullanılabilirliğinin sitotoksik ve histopatalojik deneylerle araştırılması amaçlanmıştır. In vitro deneyde 1300 kD molekül ağırlığında ve % 2, % 1, % 0.5, % 0.2, % 0.1 ve % 0.05 olmak üzere altı farklı konsantrasyondaki hyaluronik asit örneklerinin L-929 fare fibroblastı hücre kültürüne eklenmesi sonrası 24, 48 ve 72 saat sonraki sitotoksik etkileri ışık mikroskobunda değerlendirilmiştir. Vasat değişimi sonrası hücrelerde gözlenen proliferasyon sitotoksik etkinin geriye dönüşümlü olduğunu göstermiştir. Yapılan istatistiksel analiz, doz arttıkça toksik etkinin de arttığım ortaya koymuştur. Yapılan histopatalojik deneyde ise, iki farklı konsantrasyondaki hyaluronik asit solüsyonu 3 köpeğin toplam 84 diş pulpasında oluşturulan mekanik perferasyonlar üzerinde denenmiş ve 1 hafta, 1 ay ve 3 aylık periyodlar sonunda yapılan ışık mikroskobu incelenmesinde, %2 ve %0.2 konsantrasyondaki hyaluronik asitin kontrol grupları olan inert teflon maddesinden daha olumlu ve kalsiyum hidroksite benzer şekilde tamir dentini oluşturduğu tespit edilmiştir. Uygulanan düşük dozdaki hyaluronik asitin, hücre üzerine olan biyolojik etkileri nedeniyle pulpa tamiri için elverişli bir ortam sağladığı sonucuna varılmıştır. 94 SUMMARY In this study, it is aimed to evaluate the effect of hyaluronic acid used as a capping agent on dental pulp by cytotoxic and hystopathological experiments as this molecule has major roles in wound healing, cell migration, proliferation and differentiation. In the in vitro study, 2 %,1 %,0.5 %, 0.2 %, 0.1 %, 0.05 % six different concentrations of hyaluronic acid which had a molecular weight of 1300 kD was added to the L-929 mouse fibroblast cell culture and the cytotoxic effect was evaluated by light microscope after 24, 48 and 72 hours. The cell proliferation after the change of medium showed that the cytotoxic effect was reversible. The statistical analysis revealed that, as the concentration increases, the toxic effect increases also. In the histopathological experiment two different concentrations of hyaluronic acid were used as a capping material on 3 dogs' 84 dental pulps which were mechanically exposed. After 1 week, 1 month, 3 months of follow-up periods, light microscobic evaluation showed that, 2 % and 0.2 % concentrations of the hyaluronic acid showed better healing process than the inert teflon material and formed reparative dentine similar to the calcium hydroxide which were used in control groups. It is concluded that lower concentrations of hyaluronic acid may provide as suitable environment for pulp regeneration by its biological effects on cell behavior.
