Abstract
Ill THE INVEST IGA TION OF INHIBITOR Y EFFECTS OF SOME LACTOBACILLUS SPECIES UPON CLOSTRIDIUM SPECIES M.Sc. Thesis Hülya ŞİMŞEK GAZİ UNIVERSITY INSTITUTE OF SCIENCE AND TECHNOLOGY 1996 SUMMARY The antagonistic effects of Lactobacillus species on Clostridium hotulinum type B, Clostridium perfringens and Clostridium sporogenes were examined in research. Antagonistic effects were seen by well diffusion method. RCM (Reinforced Clostridial Medium) agar and BHI (Brain Heart Infusion) agar were used in this method. The antagonistic effect resulting from hydrogen peroxide which was produced by Lactabacilli was eliminated by anaerobic jar (gas-pack). In order to eliminate effect of acid 0.2% glucose was added to BHI agar and RCM agar. For the same reason, in these medium 10% CaC03 solution was used too. In this research, all Lactobacillus species showed an inhitory effect against Clostridium strains. In BHI agar all strains have formed inhibition on C. perfringens. In same medium, inhibition was observed on 8 Lb. helreficus out of 14, Lb. bulgaricus, Lb. casei ssp. tolerans and Lb. fact is against C. sporogenes. 10 out of 14 Lb. helveticus, Lb. laclis, 1 out of 2 Lb. casei ssp. tolerans, Lb. acidophilus, Lb. planfarum, Lb. casei ssp. rhamnosus and Lb. bulgaricus strains were effective upon C. hotulinum. In glucose added BHI agar, 12 Lb. helveticus out of 14, I Lb. casei ssp. tolerans out of 2, Lb. laclis, Lb. acidophilus, Lb. plantarum, Lb. casei ssp. rhamnosus and Lb. bulgaricus were formed bacteriocin-like effect on C. perfringens. All strains effected C. hotulinum. But 12 Lb. helveticus strains out of 14 and 1 Lb. casei ssp. tolerans strain out of 2 did not effect against C. sporogenes. Another strains formed inhibition. In CaCC»3 added BHI agar all strains (except Lb. casei and Lb. casei ssp. rhamnosus) formed antagonism effect on C. perfringens. 5 out of 14 Lb. helveticus effected C. sporogenes while the rest 9 /A helveticus and Lb. acidophilus did not show effect. Only Lb. acidophilus formed inhibition against ( '. hotulinum. In glucose added RCM agar all strains (except 1 Lb. case i ssp. tolerans) showed inhibitory effect against C. perfringens. All strains (except 3 Lb. helveticus) effected C. sporogenes, while only 1 Lb. helveticus strains was not effective on C. botulinum. In the end of study the substance that prevent growth of Clostridia was defined as a bacteriocin-like substance.
