Abstract
Gelisen teknoloji, artan dünya nüfusu, azalan petrol rezervleri ve maliyetinyükselmesiyle ülke ekonomisine dogrudan ve büyük katkı saglaması, ulusalkaynakların korunması ve degerlendirilmesi açısından fermentasyonla alkolüretimi güncellik kazanmıstır. Bu nedenle etanol verimini arttıracakfermentasyon kosullarının belirlenmesi önem kazanmıstır. Bu çalısmadaSaccharomyces cerevisiae mayası kullanılarak yapay melastan etanolüretiminde, fermentasyon ortamına eklenen maddelerin, etanol derisimi, etanolyüzdesi ve üretim hızı üzerine etkisi incelenmistir. Çalısma üç asamadayapılmıstır. lk asamada ortama etanol üretimi üzerine olumlu etkileriliteratürde belirlenmis olan zeolit NaY, montmorillonit(MNT) ve klinoptilolit(KLN) baslangıç pH degeri 4,5 ve degisen seker derisimi 200 ve 300 gL-1degerlerinde ortama ilave edilmistir. Bu sartlar altında ortama NaYeklenmesiyle 200 gL-1 seker ve 15 gL-1 NaY içeren ortamda üretim hızıartmıstır. Ortamda 10 gL-1 MNT bulunmasıyla, etanol derisimi artmıs, aynızamanda 200 gL-1 seker içeren ortamda maksimum derisime ulasmak için geçensüre azalmıs ve etanol üretim hızı % 16 oranında artmıstır. 5 gL-1 KLN varlıgıetanol üretim hızını %50 oranında artmıstır. Çalısmanın ikinci asamasındafermentasyon ortamına aktif karbon, bentonit ve talk ilave edilmis, deneyler100 gL-1 seker varlıgında yapılmıstır. 10 gL-1 bentonit varlıgında teorik verimv% 6 oranında artmıstır. 10 gL-1 aktif karbon varlıgında en yüksek verimeulasmak için geçen süre azalmıstır. Talk ilavesi fermentasyonu olumsuz yöndeetkilemistir. Çalısmanın üçüncü bölümünde, Box Wilson deneysel tasarımyöntemi kullanılarak Na, Ca ve Mg iyonlarının etanol fermentasyonunda kendiaralarındaki etkilesiminin etanol verimine etkisi arastırılmıstır. Çalısılanderisim aralıklarında Na'un en düsük (930mgL-1) , Ca'un orta (1515 mgL-1) veMg'un orta (128 mgL-1) kodlanmıs degerlerinde en yüksek etanol derisimineulasılmıstır. Na'un tek basına etkili oldugu, Mg ve Ca iyonlarının birbirinietkiledigi sonucuna varılmıstır. The production of ethanol with the method of fermentation has gained greatimpetus due to advancement in technology, the rapid increase in the worldpopulation, the run out of petroleum fuels reserves in crust of earth, therepercussions of the skyrocketing of costs in national economy and protection anduse of the national resources. That is why, the determination of conditions that willincrease the output efficiency of ethanol is necessary for reproduction anddevelopment of microorganisms and the efforts being made on this subject havegreat importance. The purpose of this study is to investigate the effects of materialsadded to fermentation medium that is created by Saccharomyces cerevisiae, onethanol production, ethanol concentration, production rate and pH. The study isexecuted in three steps. In the first step NaY, MNT and KLN are added to mediumwith an initial pH value of 4,5. Under these circumstances, adding 15 gL-1 NaY tofermentation medium containing 200 gL-1 sugar, production rate increased. Whenthe medium contains 10 gL-1MNT, the ethanol concentration reached to maximumvalue in the existence of 200 gL-1sugar, on the otherhand the time required toreach to maximum concentration is decreased. The ethanol production rate isincreased by 16 percent. The presence of 5 gL-1KLN did not have any influence onethanol concentration, but the production rate is raised resulting in approximately%50. In the second stage, active carbon, bentonite and talk are inserted intoviifermentation medium. The experiments were made with a fermentation mediumwhich contains 100 gL-1sugar. Presence of 10 gL-1bentonite, yield increased %6,when 10 gL-1 active carbon exists time that was required for reaching to maximumyield decreased. The supplementation of talk adversely affected the fermentation.In the third stage, we investigated the effect of change of Na, Ca and Mg ions onethanol yield. The maximum ethanol concentration level was reached at the lowestNa concentration, the middle Ca concentration and the middle Mg concentrationfor the coded values of 931mgL-1, 1515 mgL-1, 128 mgL-1respectively.
