Abstract
Difteri, Corynebacterium diphtheriae bakterisi tarafından üretilen toksin ile oluşan bir üst solunum yolu hastalığıdır. Dünya Sağlık Örgütü' nün 2013 yılına ait verilerine bakıldığında; difteri hastalığının hala dünyanın birçok bölgesinde meydana geldiği görülmektedir. Ülkemizde bu hastalığın görülme oranı azalmış olsa da; yetişkin popülasyonda duyarlılığın yüksek olması, aşılama hedeflerine ülkenin her yerinde istenilen oranlarda ulaşılamaması ve koruma amaçlı olarak kullanılan aşıların yurt dışından ithal edilmesi gibi nedenler, difteri hastalığını potansiyel bir sorun olarak karşımıza çıkarmaktadır. Difterinin klinik olarak tanısı kültür yöntemiyle yapılmakta ve bu da belirli bir zamanı kapsamaktadır. Bu durumda erken tanı ve tedavi hastalığın kontrolü için oldukça önemlidir. Monoklonal antikor temelli tanı kitleri oldukça hızlı ve güvenilir sonuçlar vermektedir. Bu çalışmada difteri toksinine karşı monoklonal antikor üretilmesi ve üretilen antikorun karakterizasyon çalışmalarının yapılması amaçlanmıştır. Bu amaç doğrultusunda; antijen olarak kullanılan difteri toksoidi, belirli dozlarda ve belirli zaman aralıklarıyla 6-8 haftalık Balb/C türü farelere intraperitonal yolla enjekte edildi. Farelerin antikor titreleri dolaylı ELISA yöntemiyle test edildi ve en iyi immün yanıtı veren fare seçilerek füzyona alındı. Polietilen glikol (PEG) varlığında, füzyona alınan farenin dalak hücreleri ve F0 myeloma hücreleri kullanılarak füzyon gerçekleştirildi. Spesifik antikor üreten hibridoma hücreleri (2F5, 5A5, 5A6) dolaylı ELISA ile belirlendikten sonra bu hücreler in vitro olarak (bulundukları besi ortamında) çoğaltıldı. En yüksek antikor yanıtı veren 5A5 klonu için ultrafiltrasyon ve jel filtrasyon kromatografi metotları ile saflaştırma işlemi gerçekleştirildi. Elde edilen monoklonal antikorun immünglobulin tipinin IgG1 ve hafif zincirinin ise κ (kappa) olduğu belirlendi. Western blot analiziyle 5A5 monoklonal antikorunun, toksinin hem A hem de B fragmentine karşı oluştuğu belirlendi. İn vitro nötralizasyon testi ile ultrafiltratın flokülasyon sınırı 40 Lf/ml, 25 nolu fraksiyonun ise 5 Lf/ml olarak bulundu. Diphtheria, which is caused by diphtheria toxin from Corynebacterium diphtheria is an upper respiratory tract disease. Looking at the World Health Organization (WHO) data in 2013, it is observed that diphtheria still occurs in many parts of the world. Even though the incidence of this disease has decreased in our country, the reasons such as the high sensitivity in the adult population, not achieving immunization targets at the desired rate in all over the country and the vaccines used for protection to be imported from abroad, etc. emerges diphtheria as a potential problem. The clinical diagnosis of diphtheria is made by culture method, and that includes a specific time. In this case, the early diagnosis and treatment is very important for disease control. Monoclonal antibody based diagnostic kits provide very fast and reliable results. In this study, the production of monoclonal antibody against diphtheria toxin and the characterization of the produced antibody are intended to work. For this purpose, diphtheria toxoid used as an antigen were intraperitoneally injected to 6-8 weeks old Balb/C mice at specific doses and time intervals. The antibody titers of mice were determined by indirect ELISA, and the best immune response in mice were selected for the fusion The fusion was carried out using the mouse spleen cell and the F0 myeloma cells in the presence of polyethylene glycol (PEG). After determining the hybridoma cells producing specific antibodies (2F5, 5A5, 5A6) by indirect ELISA, these cells were amplified in vitro (in which the cultivation medium). For 5A5 clone giving the highest antibody response, the purification process was performed using the methods of ultrafiltration and gel filtration chromatography. The immunoglobulin type and the light chain type of the obtained monoclonal antibody were determined as IgG1 and κ (kappa), respectively. 5A5 monoclonal antibody directed against both A and B fragments of diphtheria toxin were determined by Western blot analysis. With in vitro neutralization test, the limit of flocculation and 25 numbered fraction was found as 40 Lf/ml and 5 Lf/ml, respectively.
