Abstract
Bu çalışmada, 6-aminopenisilanik asit (6-APA) ve 7-aminosefalosporik asit (7-ACA) bileşiklerinden yola çıkılarak yeni β-laktam antibiyotiklerin sentezlenmesi amaçlandı. Bunun için, 5-nitrosalisilaldehit ile üç adet yüksek (büyük mol kütleli) amino asitten (4-aminobütanoik asit, 5-aminopentanoik asit ve 6-aminohekzanoik asit), sodyum bikarbonat varlığında, monosodyum tuzu formunda Schiff bazları (HLNa) sentezlendi. Monosodyum-Schiff bazlarının uygun pH'ya asitlendirilmesi sonucunda nötral formda Schiff bazları (H2L) elde edildi. 6-APA ve 7-ACA bileşikleri ile nötral-Schiff bazları kullanılarak altı adet β-laktam türevi (HL-APA, HL-ACA) elde edildi. Monosodyum-Schiff bazlarının üç adet Pd(II) kompleksi (L-Pd) sentezlendi. Sentezlenen bileşiklerin yapısı element analizi, IR, 1H/13C-NMR, UV-GB, toz XRD, XPS, kütle spektrumları TGA, manyetik duyarlılık ve iletkenlik ölçümleri ile aydınlatıldı. Monomerik komplekslerin kare düzlem yapıda olduğu ve iletkenlik göstermediği belirlendi. Bütün bileşiklerin antibakteriyal aktiviteleri mikrodilüsyon metodu ile üç adet gram negatif bakteri (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Acinetobacter baumannii RSKK 02026), üç adet gram pozitif bakteri (Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 07005, Bacillus subtilis ATCC 6633) ve bu bakterilerin ilaç dirençli izolatlarına karşı belirlendi. Antifungal aktivite için iki adet maya (Candida albicans ATCC 10231 ve C. krusei ATCC 6258) kullanıldı. Schiff bazları ile Pd(II) komplekslerinin iki adet kanser (HeLa ve MCF-7) ve bir adet normal (HEK293) insan hücre dizisi üzerindeki sitotoksik etkileri araştırıldı. Schiff bazlarının antioksidan aktiviteleri 2,2'-difenil-1-pikrilhidrazil (DPPH) serbest radikal giderim yöntemi ile MeOH ve/veya sulu ortamlarda belirlendi. In this work, it is aimed to synthesis of new β-lactam antibiotics derived from 6-aminopenicillanic acid (6-APA) and 7-aminocephalosporanic acid (7-ACA). For this, 5-nitrosalicylaldehyde was reacted with three higher (high molecular weight) amino acids (4-aminobutanoic acid, 5-aminopentanoic acid and 6-aminohexanoic acid) in the presence of sodium bicarbonate and then Schiff bases were synthesized as monosodium salt form (HL-Na). Neutral-Schiff bases (LH2) were obtained from the monosodium-Schiff bases by adjusting to a suitable pH. And then, β-lactam derivatives (HL-APA, HL-ACA) were produced by using 6-APA, 7-ACA and the neutral-Schiff bases. Three Pd(II) complexes (L-Pd) were synthesized by using monosodium-Schiff bases. The synthesized compounds were characterized by elemental analysis, IR, 1H/13C NMR, UV-vis, powder XRD, XPS, mass spectra, TGA, conductivity and magnetic susceptibility measurements. All the monomeric complexes were found to have square planar geometry and nonelectrolytic nature. Antibacterial activities of all the compounds were screened by microdilution method against three gram negative bacteria (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Acinetobacter baumannii RSKK 02026), three gram positive bacteria (Staphylococcus aureus ATCC 25923, Enterococcus faecalis ATCC 07005, Bacillus subtilis ATCC 6633) ve and their drug-resistant isolates. Two fungi (Candida albicans and C. krusei) were used for antifungal activity. Cytotoxic effects of Schiff bases and Pd(II) complexes were investigated against cancer human cell (HeLa and MCF-7) and normal HEK293 cell. Schiff bases were evaluated for antioxidant activities by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method in both methanolic media and/or aqueous solutions.
