Abstract
Bu tez çalışması genel olarak 4 aşamadan oluşmaktadır. İlk aşamada, floresans özellik gösteren bir perilen türevi prob sentezlenmiş ve FT-IR, 1H NMR ve kütle spektroskopisi gibi spektroskopik yöntemler kullanılarak karakterizasyonu gerçekleştirilmiştir.Çalışmanın ikinci aşamasında, sentezlenen molekülün metal iyonlarına karşı seçicilik ve duyarlılığı, floresans spektroskopisi kullanılarak incelenmiştir. Sentezlenen perilen türevi probun Hg2+ iyonuna duyarlı olduğu gözlenmiştir. Ayrıca probun cevap süresi, tayin limiti ve seçicilik çalışmaları yapılmıştır. Çalışmanın üçüncü kısmında ise sentezlenen molekül ve Hg2+ iyonu ile yapmış olduğu kompleksin teorik hesaplamaları yapılmıştır. Hesaplamalarda Gaussian 09W programı ve yoğunluk fonksiyonel teorisi (DFT) yönteminin B3LYP fonksiyonu ile 6-31G(d) ve LANL2DZ baz seti kullanılarak moleküllerin geometrik optimize yapılarının yanında HOMO ve LUMO orbitalleri ve enerjileri hesaplanmıştır.Çalışmanın son aşamasında ise geliştirilen floresans probun HepG2 karaciğer kanser hücrelerinde görüntüleme çalışması gerçekleştirilmiştir. Sonuç olarak, sentezlenen perilen türevli prob, Hg2+ metal iyonuna karşı yüksek seçicilik ve duyarlılık göstermiştir. Ayrıca hücre içi floresans görüntüleme çalışmalarında başarılı sonuçlar elde edilmiştir. This thesis consists of 4 stages. In the first stage, the perylene derivative probe which showed fluorescence behaviour was synthesized and characterized by using FT-IR, 1H NMR and mass spectroscopy.In the second stage of these study, the selectivity and sensitivity of the synthesized molecule against metal ions were investigated by using fluorescence spectroscopy. The synthesized perylene derivative probe was observed to be sensitive to Hg2+ ion. Besides, response time, detection limit and selectivity studies of the probe were performed.In the third part of the study, the theoretical calculations of the synthesized molecule andits Hg2+ ion complex were made. In the calculations, HOMO and LUMO orbitals and were calculated by using Gaussian 09W program and density functional theory (DFT) method with B3LYP function 6-31G (d) and LANL2DZ basis set.In the last part, imaging studies of the developed fluorescence sensor in the liver cancer cell HepG2 was performed. As a result, the developed perylene derivative probe showed high selectivity and sensitivity to Hg2+metal ion. Also, successful results have been obtained in intracellular fluorescence imaging studies.
