Abstract
ÖZET Sağlıklı insan periferik kan hemopoietik stem ve progenitor hücrelerinin, 10 günlük in vitro likit hücre kültüründe, matür hücrelere diferansiasyonu üzerine multi-CSF, GM-CSF ve Epo'nun etkilerini ince ledik. Multi-CSF olarak laboratuvarımızda hazırladığımız PHA-LCM'i kullandık. Sonuçlarımızı maddeliyecek olursak; Sitokinlerin ve kombinasyonlarının azalan yanıt sırasına göre (1-4): 1- Total WBC progenitörleri için; PHA - LCM + GM - CSF > PHA -LCM + Epo > PFİA - LCM > PHA - LCM + GM - CSF + Epo > GM -CSF = GM - CSF + Epo = Epo. 2- Nötroffl progenitörleri için; PHA - LCM + Epo > PHA - LCM > PHA - LCM + GM - CSF + Epo > Epo > PHA - LCM + GM - CSF > GM - CSF + Epo = GM - CSF 3- Bazofil progenitörleri için; PHA - LCM > PHA - LCM + GM - CSF > PHA - LCM + Epo > PHA - LCM + GM - CSF + Epo > Epo= GM - CSF + Epo = GM - CSF 4- Monosit progenitörleri için; PHA - LCM + GM + GM - CSF + Epo > PHA - LCM + GM - CSF > PHA - LCM > PHA - LCM + Epo > GM - CSF + Epo = Epo = GM - CSF. 5 ve 6'da sitokinlerin ve kombinasyonların azalan supresyon oluşturma etkilerine göre; 675- Nötrofîl progenitörleri için; PHA - LCM + GM - CSF > PHA - LCM + GM - CSF + Epo > PHA - LCM + Epo > GM - CSF + Epo = PHA - LCM = Epo = GM - CSF 6- Lenfosit progenitörleri için; PHA - LCM > PHA - LCM + GM - CSF + Epo > PHA - LCM + GM - CSF > PHA- LCM + Epo > GM - CSF + Epo = Epo = GM - CSF 68 SUM M ARY We examined the effect of multi - CSF, GM - CSF and Epo on the dif ferentiation of healthy human. peripheric blood hemopoietic stem and progenitor cells in to their mature types in a 10 day - in vitro liquid cell culture. As multi - CSF we used PHA - LCM which had been prepared in our laboratory. Our results are listed as follows: Numbers 1 through 4 showed dec reasing responsiveness among cytokines and their combinations. 1- For total WBC progenitors; PHA - LCM + GM - CSF > PHA -LCM + Epo > PHA - LCM > PHA - LCM + GM - CSF + Epo > GM -CSF = GM - CSF + Epo = Epo. 2- For platelet progenitors; PHA - LCM + Epo > PHA - LCM > PHA - LCM + GM - CSF + Epo > Epo > PHA - LCM + GM - CSF > GM - CSF + Epo = GM - CSF 3- For basophil progenitors; PHA - LCM > PHA - LCM + GM - CSF > PHA - LCM + Epo > PHA - LCM + GM - CSF + Epo > Epo= GM - CSF + Epo = GM - CSF 4- For monocyte progenitors; PHA - LCM + GM + GM - CSF + Epo > PHA - LCM + GM - CSF > PHA - LCM > PHA - LCM + Epo > GM - CSF + Epo = Epo = GM - CSF. Numbers 5 and 6 showed decreasing suppression among cytokines and their combinations. 695- For neutrophil progenitors; PHA - LCM + GM - CSF > PHA - LCM + GM - CSF + Epo > PHA - LCM + Epo > GM - CSF + Epo = PHA - LCM = Epo = GM - CSF 6- For lymphocyte progenitors; PHA - LCM > PHA - LCM + GM - CSF + Epo > PHA - LCM + GM - CSF > PHA - LCM + Epo > GM - CSF + Epo = Epo = GM - CSF 70
