The cytotoxic, apoptotic, genotoxic and oxidative effects of carbonic anhydrase 9 inhibitor on cervical cancer cells

Abstract
Serviks kanseri kadınlarda en sık görülen kanser türlerinden biridir. Kemoterapi, kanser tedavisinde kullanılan yaygın bir terapi olarak öne çıkmaktadır. Karbonik anhidraz IX (CAIX) kanser tedavisinde özellikle cazip bir hedeftir; çünkü normal dokularda sınırlı olarak ifade edilirken çok çeşitli kanserlerde aşırı eksprese olur. Bu nedenle, CAIX inhibitörü olarak sülfonamid türevinin antikanser özelliklerini CAIX ekspresyonu pozitif olan serviks kanser hücreleri (HeLa) ve negatif olan normal hücrelerde (PNT1-A) araştırmayı planladık. Çalışmanın amacı, servikal kanser (CC) hücre dizisi (HeLa) üzerinde yeni sentezlenmiş sülfonamid türevinin (S-1) bazı moleküler mekanizmalarını değerlendirmektir. Hücre canlılığı WST-1 analizi ile saptanmış ve daha sonra IC50 değeri değerlendirilmiş, apoptozis ve hücre döngüsü indüksiyonu flow sitometri cihazıyla ölçülmüş, intraselüler serbest radikal indüksiyonu reaktif oksijen türü (ROS) analizleri ile gerçekleştirilmiştir. S-1'in hücre canlılığını önemli ölçüde düşürdüğünü, ROS üretimini yukarı regüle eden önemli apoptoz belirtileri ortaya çıkardığını bulduk. Hücre hayatta kalma oranında görülen bu düşüş yüksek seviyedeki ROS ve apoptoza bağlanabilir ki bunun aynı zamanda hücre döngüsünü durdurduğu da saptanmıştır. Bulgularımız yeni sentezlenmiş sülfonamid türevi S-1'in Hela'da PNT1-A'ya göre daha etkili olduğunu gösterdi. Bu nedenle türev bileşik S-1, servikal kanserde bir anti-kanser potansiyeli olarak kullanılabilir. Sonuçlar, S-1'in serviks kanseri hücrelerinin apoptozunu indükleyebildiğini ve gelecekteki kanser çalışmalarında olası bir aday olduğunu göstermiştir. Cervical cancer is one of the most common types of cancer in women. Chemotherapy stands out as an extensive therapy used in the treatment of cancer. Carbonic anhydrase IX (CAIX) is an especially attractive target for cancer therapy, because of the overexpressed in a wide variety of cancer, while limited way in normal tissues. For this reason, we planned to investigate the anticancer properties of the sulfonamide derivative as a CAIX inhibitor on cervical cancer cells (HeLa) positive for CAIX expression and normal cells (PNT1-A) negative for CAIX expression. The purpose of the study to evaluate of some molecular mechanism of novel synthesized sulfonamide derivative (S-1) on cervical cancer (CC) cell line (Hela). In this study, we examined the cytotoxic, apoptosis, genotoxic and oxidative stress activity of S-1 on Hela and normal prostate epithelium cell line (PNT1-A). Cell viability determined by WST-1 assay and then IC50 value evaluated, apoptosis and cell cycle induction measured by flow cytometric analyzer, intracellular free radical induction performed by reactive oxygen species (ROS) analyses. We found that S-1 induces significantly reduce cell viability, caused significant signs of apoptosis level, also up-regulating ROS production. This decrease in cell survival rate can be attributed to the high level of ROS and apoptosis, which has also been shown to arrest cell cycle. Our findings indicated that novel synthesized sulfonamide derivative S-1 is more effective on Hela than PNT1-A. Therefore, derivative compound S-1 might be used as an anti-cancer compound on cervical cancer. The results showed that the S-1 is able to induce the apoptosis of cervical cancer cells and is a possible candidate for future cancer studies.
