Üridinin yenidoğan hiperoksik beyin hasarı modelinde serbest oksijen radikalleri üzerine etkisinin araştırılması

Abstract
Preterm yenidoğanlara neonatal yoğun bakım ünitelerinde, resüsitasyon,pulmoner hipertansiyon ve solunum sıkıntısı gibi nedenlerden dolayı oksijen tedavisiuygulanmaktadır.Term ve preterm yenidoğanlar, endojen radikal süpürücü sistemlerinin tamolarak olgunlaşmamış olması sebebiyle reaktif oksijen türlerinin neden olduğubozulmalara duyarlıdırlar. Reaktif oksijen türleri, fizyolojik olarak antioksidansistemler aracılığı ile düzenlenmektedir. Reaktif oksijen türleri ve antioksidanarasındaki dengenin bozulması DNA ve mitokondriyal membran, apoptoz ve hücreselfonksiyon bozukluğuna bağlı olarak oksidatif strese neden olabilir. Oksidatif stresinhiperoksiyi takiben akciğer, retina ve muhtemelen genel doku hasarından sorumluolduğu kabul edilmiştir. Bu çalışmanın amacı, yenidoğan sıçan beyninde hiperoksi uygulamasısonrasında oluşan oksidatif stresin neden olduğu bozulmalara karşı üridinin,antioksidan sistem aracılığıyla olası tedavi etkinliğini araştırmaktır. Çalışmamızda 53adet neonatal Spraque-Dawley cinsi sıçan doğumdan 6 gün sonra randomize olarak 5gruba ayrılmıştır. Oda havasındaki sıçanlara 6. gün tek doz intraperitoneal serumfizyolojik (%0,9 NaCl) veya diğer gruplara hiperoksi maruziyetinden 15 dakika öncetek doz 100 mg/kg, 300 mg/kg, 500 mg/kg dozunda üridin intraperitoneal (i.p.)enjeksiyonu yapılmıştır. 48 saat boyunca oda havasında (%21 O2) tutulmuş veyahiperoksiye (%80 O2) maruz bırakılan sıçanlar takiben dekapite edilerek tüm beyinhomojenatlarından Western Blot yöntemiyle DJ-1 proteininin analizi veMalondialdehit (MDA), Myeloperoksidaz (MPO), Süperoksit Dismutaz (SOD),Glutatyon Peroksidaz (GSH-Px) enzimlerinin biyokimyasal analizleri yapılmıştır. Oxygen therapy is applied to preterm neonates in neonatal intensive care unitsfor reasons such as resuscitation, pulmonary hypertension and respiratory distress. Term and preterm neonates are susceptible to deterioration caused by reactiveoxygen species because their endogenous radical scavenging systems are not fullymature. Reactive oxygen species are regulated physiologically by antioxidant systems.Disruption of the balance between reactive oxygen species and antioxidants may causeoxidative stress due to DNA and mitochondrial membrane, apoptosis and cellulardysfunction. The degradation of balance between reactive oxygen species andantioxidant may cause oxidative stress due to DNA and mitochondrial membrane,apoptosis and cellular dysfunction. Oxidative stress has been recognized to beresponsible for lung, retina and possibly general tissue damage following hyperoxia. The aim of this study was to investigate the possible treatment efficiency ofuridine through antioxidant system against the deterioration caused by oxidative stressafter hyperoxia application in pups brain. In our study, 53 neonatal Spraque-Dawleyrats were randomly divided into 5 groups 6 days after birth. Rats in room air wereinjected intraperitoneally (i.p.) a single dose of either saline (0.9% NaCl) or uridine100mg/kg, 300 mg/kg, 500 mg/kg 15 minutes prior to hyperoxia exposure to on day 6. Ratpups which were kept either at room air (21% O2) or exposed to hyperoxia (80% O2)for 48 hours, were subsequent to decapitated. The homogenates obtained from thewhole brain were analyzed by Western blotting DJ-1 protein, and the biochemicalanalysis of Malondialdehyde (MDA), Myeloperoxidase (MPO), SuperoxideDismutase (SOD), Glutathione Peroxidase (GSH-Px) enzymes were conducted.
