Abstract
Akut lösemiler olgun hücrelerin azalması ve lökosit prekürsörlerinin (lösemik blastlar) birikimi ile karakterizedir. Bu çalışmada HL-60 insan Akut Promyelositik Lösemi (APL) hücre kültüründe Sorafenib ve Lityum Klorür (LiCl)'ün etkilerini araştırmak amaçlanmıştır. Deneylerde Sorafenib ve LiCl ilaçları tek tek ve birlikte, insan kökenli HL-60 APL hücrelerinden oluşan kültürde 72 saat boyunca denenmiştir. Tümör hücre sayıları sayım kamarasıyla, apoptoz oranları akım sitometri yöntemiyle değerlendirilmiştir. Hücre kültüründe hücrelerde meydana gelen değişiklikler geçirimli elektron mikroskobu ile araştırılmıştır. İlaçların etkileri hangi yoldan oluşturduklarını saptamak için ELISA yöntemi ile kaspaz-3 aktivitesi, fosforile GSK-3ß, fosforile AKT, total AKT, fosforile p38, total p38, fosforile ERK, total ERK, fosforile I?B?, total I?B? , fosforile c-jun, total c-jun, fosforile STAT3 ve total STAT3 seviyelerine bakıldı. Sorafenib ve LiCl'ün tek tek ve birlikte verilmesi durumunda HL-60 insan hücre kültüründe kontrol grubuna göre istatistiksel olarak anlamlı derecede hücre sayısını azalttığı ve apoptoz oranını artırdığı tespit edilmiştir. Acute leukemia is characterisized with decreasing mature cells and aggregation of leucocyte precursors. In this study we aimed to investigate the effects of Sorafenib and Lithium Chloride (LiCl) on HL-60 human Acute Promyelocytic Leukemia (APL) cell line. The HL-60 APL cell cultures are subjected to Sorafenib and LiCl separately and in combination for 72 hours during experiments. Tumor cells are counted under Cell Count Hemocytometer and the apoptose ratios are investigated with flow cytometry method. The changes in the cells in cell cultures are evaulated by transmission electron microscopy. To determine the possible the mechanism of action of the drugs, the caspase 3 actvities, phosphorylated GSK-3ß, phosphorylated AKT, total AKT, phosphorylated p38, total p38, phosphorylated ERK, total ERK, phosphorylated I?B?, total I?B? , phosphorylated c-jun, total c-jun, phosphorylated STAT3 ve total STAT3 levels are investigated with ELISA method. Separately and in combination Sorafenib and LiCl decreased the number of cells and increased the apoptotic ratio in HL-60 APL cell line in a statistically significant manner when compared with the control group.
