Glutamat nörotoksisitesi oluşturulan sıçanlarda metforminin olası nöroprotektif etkisinde glukojen sentaz kinaz-3-betanın rolü

Abstract
Giriş : Glutamat santral sinir sisteminde en yaygın bulunan eksitatorik nörotransmitterdir ve Alzheimer hastalığı , iskemik hasar, amiyotrofik lateral skleroz gibi hastlıkların patogenezinde yer almaktadır. Metforminin santral sinir sistemi üzerinde koruyucu etkilerinin olduğu ileri sürülmektedir. Bu etkilerinin temel mekanizmaları henüz açıklığa kavuşmamıştır.Glukojen sentaz kinaz-3 beta (GSK-3β)'nın inhibisyonuyla metforminin etkilerinin birbirine benzerlik göstermesi metforminin söz konusu etkilerini GSK-3β modülasyonu ile yapabileceğini düşündürmüştür. Diğer taraftan çinko, GSK-3β üzerinde inhibisyon yaratan önemli bir eser elementtir.Amaç : Araştırmamızda, sıçanlarda glutamat ile oluşturulan nörotoksisiteye metforminin nörotrofik etkisi ve olası nörotrofik etkide GSK-3β'nın ve çinkonun rolü araştırıldı.Gereç ve Yöntem : 40 adet erişkin erkek sıçan 5 gruba ayrıldı; Kontrol , Glutamat, Metformin+Glutamat, Çinko Eksikliği+Glutamat, Çinko Eksikliği+Metformin+Glutamat. Metformin 34 gün süreyle verildi. 35. gün D-glutamik asit uygulandı.Takiben 3 gün sonra anesteziye sıçanlardan alınan beyin dokusu örneklerinde nörodejenerasyon histopatolojik; nörorejenerasyon immünohistokimyasal olarak değerlendirildi. Bulgular kanda ve beyin dokusunda bakılan GSK-3β ve oksidatif stres parametrelerinin düzeyleriyle ilişkili olarak irdelendi.Bulgular : Çinkodan yetersiz diyetle beslenen sıçanlarda nörodejenerasyonun derecesi yüksekti. Nörodejenerasyonun gözlendiği gruplarda sıçanların kan ve beyin dokusunda GSK-3β düzeylerinde artışlar saptandı. Metformin verilen gruplarda nörodejenerasyonun azaldığı; nörorejenerasyonun arttığı gözlendi. Bu gruplarda kan ve beyin dokusunda GSK-3β ve oksidatif stres parametrelerinde azalmalar, anti-oksidan parametrelerde artışlar gözlendi. Metforminin yetersiz çinko diyeti ile beslenen sıçanlarda protektif ve rejeneratif etkilerinin, normal diyet ile beslenen sıçanlardan daha az olduğu belirlendi.Sonuç : Metformin nöroprotektif ve nörorejeneratif etkilerini, GSK-3β 'nin ekspresyonunu ve/veya aktivasyonunu çeşitli yolaklarla indirekt ve /veya direkt olarak azaltarak gösterebilir. Metforminin nöroprotektif ve nörorejeneratif etkisinde çinko iyonu önemli rol oynayabilir. Glutamate is the most common excitatory neurotransmitter in the central nervous system(CNS) and involved in the pathogenesis of diseases such as Alzheimer's disease, ischemic injury, amyotrophic lateral sclerosis.Metformin has been suggested to have protective effects on the CNS. Mechanisms of these effects have not yet been clarified.The similarity of the effects of metformin with the inhibition of GSK-3β suggests that metformin may moduate GSK-3β. On the other hand, zinc is an important trace element that inhibits GSK-3β.In our study, the neurotrophic effect of metformin on glutamate-induced neurotoxicity in rats and the role of GSK-3β and zinc in these neurotrophic effects were investigated.Forty adult male rats were divided into 5 groups; Control, Glutamate, Metformin+Glutamate, Zinc Deficiency+Glutamate, Zinc Deficiency+Metformin+Glutamate. Metformin was given for 34 days. D-glutamic acid was administered on the 35th day. Neurodegeneration was examined histopathologically and neuroregeneration was evaluated immunohistochemically in brain tissue samples taken from anesthetic rats. The findings were examined in relation to levels of GSK-3β and oxidative stress parameters observed in brain tissue and blood.Neurodegeneration was higher in rats fed with inadequate zinc diet. GSK-3β levels were increased in groups with neurodegeneration. Decreased neurodegeneration and increased neuroregeneration; decreased GSK-3β and oxidative stress parameters, increased antioxidant parameters was obsereved in groups treated with metformin. Metformin had less protective and regenerative effects in rats fed with inadequate zinc diet.Metformin may demonstrate neuroprotective and neuroregenerative effects by reducing expression of GSK-3β. Zinc may play an important role in neuroprotective and neuroregenerative effects of metformin.
