Abstract
SNP'ler, tahmini her 1000 bazda bir sıklıkta ve en çok bulunan DNA belirteçleridir. SNP'ler, genomun hem kodlayan, hem de kodlamayan bölgelerinde meydana gelebilip, konservatif olmayan amino asit veya gen regülasyonu değişimlerine neden olabilirler. Tek aşamalı, PZRtemelli, üniversal analiz metodu olan Amplifluor® SNP Genotipleme Sistemi, SNP genotipleme analizini oldukça kolaylaştırmaktadır. Bu sistem oldukça duyarlı, homojen bir floresan analiz yöntemidir. Allel spesifik PCR ile analiz edilebilecek her türlü biyolojik örneğe adapte edilebilmektedir.Adli olguların aydınlatılabilmesi, sıklıkla eser miktardaki biyolojik materyalin DNA düzeyinde incelenmesini gerektirmektedir. Bu tür örneklerde yeterli miktarda nükleer DNA'ya ulaşmak her zaman mümkün olamamaktadır. Nükleer DNA ile incelenmesi çok zor olan eski kemikler, ileri derecede degrade olmuş biyolojik materyal veya tek ve köksüz kıl örneklerinde mitokondriyal DNA ile büyük başarı elde edilmektedir. Maternal kalıtımı nedeni ile mtDNA akrabalık ilişkilerinin belirlenmesinde de faydalıdır. Bu tezin amacı, bir mtDNA genotipleme metodu standardize etmek ve Türk popülasyonunun mutasyonları ile ilgili olarak veri elde etmekti.mtDNA 7025. SNP bölgesi, Real-Time PCR cihazı ile genotiplendi ve haplotipler, adli amaçlı mtDNA genotiplemede Real-Time PCR'ın uygulanabilirliğini anlamak için, standart end-point PCR sonuçları ile karşılaştırılarak değerlendirildi. Bu nedenle çalışmamızda akrabalık ilişkisi olmayan 10 kişiden alınan kan örneklerinden DNA ticari bir kit ile çekitlenerek, gerek genotipleme yönteminin standardizasyonunun sağlanması, gerekse toplumumuzda var olan mutasyonlarla ilgili bir ön bilgi edinilmesi amaçlanmıştır. Hedef bölgeler Real-Time PZR cihazı ile çoğaltılmış ve ürünler, uygun bir işaretleme tekniği kullanılarak, üniversal Amplifluor floresan metodu ile genotiplenmiştir. SNPs are the most abundant DNA markers with an estimated frequency of 1 SNP in every 1000 bases. SNPs can occur in both coding and noncoding regions of the genome and can cause nonconservative amino acid changes or altered gene regulation. The development of a one step, PCR-based, universal detection method, the Amplifluor® SNPs Genotyping System, simplifies high throughput analysis for the genotyping of SNPs. The Amplifluor® SNPs Genotyping System is a highly sensitive, homogeneous fluorescent assay. It can be adapted to virtually any sample that can be analyzed by allele-specific PCR.It is not always possible to obtain enough nuclear DNA sample for the analysis of the biological samples in the purpose of forensic evaluations. The polymorphic DNA found in the mitochondria matrix exist at least a hundred copies and it makes easy to obtain results. Where it is not possible to work with nuclear DNA, for example ancint bones, any degraded biological material, hair shaft, mtDNA gives sucessful results. Besides these advantages, due to maternal inheritance of the mtDNA, it is useful for the investigation of kinship. The aim of this thesis is to standardize the method for the genotyping of the mtDNA and to obtain data about the mutations of the Turkish population.The 7025th SNP site of the mtDNA was genotyped with Real-Time PCR instrument and the haplotypes were evaluated compared to standart end-point PCR results for understanding the applicability of Real-Time PCR in genotyping mtDNA for forensic purposes. Blood samples were collected from10 unrelated individulas and mtDNA was extracted with a commercially available kit. The target regions were amplified by means of Real-Time PCR machine and then the products were genotyped by using an appropriate labelling for the universal Amplifluor fluoresence method.
