Abstract
DNA'nın polimorfik özelliklerinin değerlendirilmesi ile soy bağı tespiti yapılması adli DNA analizlerin en önemli kullanım alanlarından biridir.Gebelikle sonuçlanan cinsel saldırı olguları doğum öncesi erken dönem babalık tayinini gerekli kılmaktadır. Günümüzde bu tespit amniyosentez ve CVS gibi invaziv girişimsel tekniklerle yapılabilmektedir. Neden oldukları maternal ve fetal riskler sebebi ile bu yöntemlere adli makamlar tarafından nadiren başvurulmaktadır. Bu çalışmada; invaziv girişimsel olmayan bir teknikle doğum öncesi babalık tayini yapılması amaçlandı. Gönüllü katılımcılardan aydınlatılmış onamları alınarak temin edilen kan ve tükürük örnekleri, DNA profilleme amacı ile incelendi. Çalışmanın laboratuvar kısmı, T.C Adalet Bakanlığı Adli Tıp Kurumu Eğitim ve Bilimsel Araştırma Komisyonu'nun B.03.1.ATK.0.01.00.08/475 sayılı izni ile T.C Adalet Bakanlığı Adli Tıp Kurumu Biyoloji İhtisas Dairesi Laboratuvarlarında gerçekleştirildi. Çalışmada, hamile kadınların periferal kanlarında bulunan fetüse ait serbest fetal DNA'ların izolasyon, ultrafiltrasyon, kantifikasyon ve amplifikasyon basamakları için çeşitli ticari kitler kullanıldı. Miktar tayini aşamasında Real Time PCR tekniğinden yararlanıldı. PCR ürünlerinin tiplendirilmesinde kapiller elektroforez yöntemi kullanıldı. Elde edilen Y-STR profil sonuçları muhtemel babalarınkiler ile karşılaştırıldı. Bu çalışmada 16. haftadan itibaren fetüse ait serbest fetal DNA'ların izolasyon ile erkek fetüslerde babalık tayini yapılabileceği gösterildi. Çalışma ile maternal kandan serbest fetal DNA analizi yöntemiyle doğum öncesi babalık tayininin kullanılabilirliği belirlendi. Türkiye'de ilk kez uygulanan bu yöntemin adli sürecin hızlandırılmasına da katkıda bulunabileceği görüldü. Anne kanından fetüse ait DNA eldesi ile gerçekleştirilen testin rutinde uygulanabilirliği bu çalışma ile gösterildi. Evaluation of the polimorphic characteristics of the DNA in according to determine the family relations, is an important area of the forensic DNA analyses. Sexual assaults resulting pregnancy required prenatal paternity determination. Today this determination can be made by invasive techniques like amniocentesis and CVS. Owing the maternal and fetal risks, these tecniques are rarely used by judicial offices.In this study, we aimed to make the prenatal paternity determination using a non invasive tecnique.We took informed consents from volunteer participants for their blood and saliva samples examined according to DNA profiling. The laboratory part of the study is performed in Republic of Turkey Ministry of Justice Forensic Medicine Institution Biology Specialization Office Laboratory under the permission number B.03.1.ATK.0.01.00.08/475 of Education and Scientific Research Comission of Republic of Turkey Ministry of Justice. We used various commercial tests for isolation, ultrafixation, quantification and amplification of the free fetal DNAs in mathernal peripheral blood circulation. We used real Time PCR tecnique for the amount determination. We also used capillary electrophoresis in order to classify the PCR products. And we compared the Y-STR profile results with the probable father's results.We showed that the determination of the paternity from isolation of the circulating free fetal DNA in mother's peripheral blood is applicable from 16th week. The study stated the utilization of the prenatal paternity determination using fetal DNA analysis. This study is known to be the first in its field in Turkey, and it's also seen that this analysis can also accelerate the judical process., With this study it has been shown that, using this test of determining the fetal DNA from maternal blood is applicable in routine laboratory.
