Abstract
Adli moleküler genetikte olguların aydınlatılmasında genellikle STR/SNP genetik işaretlerinden yararlanılır. İnsersiyon-delesyon (InDel) sonucu meydana gelen kalıtımsal değişiklikler de, insan genomunda genetik işaret olarak kullanılabilmektedir. Yeni nesil genetik markırlar olarak adlandırılan InDel'ler, STR ve SNP lokuslarına alternatif ya da birlikte kullanılmasıyla kimliklendirme ve biyocoğrafik soy tayini daha başarılı yapılabilmektedir. Bu tez çalışmasında biyocoğrafik soy analizinde kullanılabilecek 46 Ancestry Informative Marker (AIM) InDel lokusu çalışılmıştır. Bu çalışmada 46 AIM InDel lokusunun Türkiye'deki gen sıklığı saptanması için 148 kişiye ait veriler genetik açıdan incelendi. Laboratuvar aşamasında, ilgili lokuslar PCR tekniği ile çoğaltıldı ve kapiler elektroforez ile görüntülenmesi sağlandı. Arlequin ver. 3.5 programı ile gen sıklıkları ve populasyonlar arası benzerlik oranı belirlendi. Fst analizi ile toplumsal tabakalaşmanın boyutu saptandı. Ayrıca birey bazında istatistiksel veri sunulması için Snipper programı kullanıldı. Türkiye populasyonuna ait genetik verilerin, 7 dünya populasyonu ile genetik açıdan karşılaştırılması Structure programı ile yapıldı. Bu tez çalışması kapsamında, 7 popülasyon (Afrika, Avrupa, Doğu Asya, Okyanusya, Amerika, Ortadoğu, Merkez Güney Asya) arasında Türkiye populasyonun yeri belirlenmiştir. Bu çalışma bulguları ile genetik açıdan Türk populasyonu ile Avrupa, Ortadoğu ve Merkez Güney Asya populasyonları arasında benzerlik görülmüştür. STR/SNP (Short Tandem Repeats / Single Nucleotide Polymorphism ) genetic markers are used for forensic identification. In order to solve many of the judicial cases. Insertion-deletion (InDel) in human genome can also be used for forensic genetic markers. Insertion/deletion polymorphism (InDeL)s, which also called ` next generation genetic variations`, has been started to used together or as an alternative to STR and SNP loci for better results in identification and estimation of the biological ancestry. In this thesis, the 46 Ancestry Informative Marker (AIM) InDel loci are studied for biogeographic ancestry analysis. In this research, we investigated the gene frequency of the 46 INDEL loci in Turkey using genetic data from 148 individuals. Related loci were amplified using PCR and electrophoresis technics. Gene frequencies and population parameters were calculated by using Arlequin ver. 3.5. The degree of the population stratification is identified by using Fst analysis. Each individual's ancestry assignments were estimated using online Snipper web tool. Structure programme is applied for population comparison of the seven reference populations and Turkish population.The results show that almost all the Turkish individuals have high European ancestry components when we test three population (Europe, East Asia, and Africa) differentiation option in the SNIPPER portal. The results unsurprisingly remain similar when the number of reference population increased with additional Americans and Oceanians. As a result of this study, the accurate inference of the unknown individual can be easily achieved for only the continental groups of Europe, Africa, and East Asia. However sub-continental differentiations, in this case differentiation of Europe versus Middle East or Central South Asia, can be more optimally differentiated by a second tier panel which includes ancestry-informative markers chosen for the target region.
