Abstract
Laktoperoksidaz (E.C. 1.11.1.7); moleküler ağırlığı yaklaşık 78 kDa olup, hem grubu ve % 8-10 karbonhidrat içeren 612 aminoasit zincirinden ibaret, tek bir polipeptid zincirinden oluşan bir enzimdir. Bu enzim, yenidoğanların sindirim sisteminden salgılanan patojen mikroorganizmalara karşı duran, tükürükte gözyaşında ve sütte bulunan bir oksidoredüktazdır. Bu çalışmada sığır laktoperoksidaz (LPO) enzimi Sepharose 4B-etilendiamin-4-izotiyosiyonat benzen sulfonamid jeli kullanılarak saflaştırılmıştır. Enzimin saflık kontrolü SDS-PAGE ile kontrol edilmiştir. Enzimin saflaştırma derecesi 212000 % verimi 0,74 olarak bulunmuştur. Çalışmamızda endüstri, ziraat ve eczacılıkta sıklıkla kullanılan kimyasalların enzim üzerine in vitro etkisi incelenmiştir. Bu amaçla çalışılan organik çözücülerin IC50 değerleri hesaplanmış ve inhibisyon etkisi gösterenleri belirlenmiştir. Organik çözücüler içinde asetofenon 1,82 M ile en fazla inhibisyon etkisi göstermiştir. 1-bütanol, sülfürik asit, nitrik asit, karbon tetra klorür, kloroform, 2- erkaptoetanol, hidrojen peroksit, asetik asit, orto fosforik asit, formik asit, indol 3-bütirik asit, sodyum hipoklorit, hidroflorik asit, kojik asit,n-pentan, dietil eter, etil asetat, amonyak, anilin, formaldehit çözücülerinin LPO enzimi üzerine aktivite etkisi gözlenmemiştir. Lactoperoxidase (E.C. 1.11.1.7); the molecular weight is about 78 kDa, and is an enzyme consisting of a single polypeptide chain consisting of 612 amino acid chains containing both the group and 8-10% carbohydrate. This enzyme is an oxidoreductase found in tears and milk in the saliva, which stands against pathogenic microorganisms released from the digestive system of newborns. In this study, bovine milk lactoperoxidase (LPO) enzyme was achieved to be purified by a affinity chromatography gel, prepared by using CNBr-activated Sepharose-4B as a matrix, ethylene amine as a spacer-arm and inhibitors, 4- isothiocyonate benzene sulfonamide as the ligand. The purity of the enzyme has been checked by SDS-PAGE. The purification value for lactoperoxidase has been found as 212000 folds with 0,74 % yield. In our study, the in vitro effects of chemicals, which are frequently used in industry, agriculture and pharmacy, on the enzyme were investigated. For this purpose, IC50 values, Ki values and types of inhibition were determined. Most inhibition by chemicals acetophenone 1,82 M was affected. 1-butanol, sulfuric acid, nitric acid, carbon tetra chloride, chloroform, 2-mercaptoethanol, hydrogen peroxide, acetic acid, ortho phosphoric acid, formic acid, indole 3-butyric acid, sodyum hypochlorite, hydrofluoric acid, kojic acid, n- pentane, diethyl ether, ethyl acetate, ammonia, aniline, formaldehyde were not effective on the enzyme.
