Thiopentalin rat karaciğer ve akciğer NADPH sitokrom P450 redüktoz, NADH sitokrom b5 redüktaz ve N-Nitrozodimetilamin demetilaz enzim aktiviteleri üzerindeki etkisi

Abstract
ÖZET Bu araştırmada, yaygın kullanılan bir anestetik madde olan thiopental'in karaciğer ve akciğer mikrozomal NADPH sitokrom P450 redüktaz, NDMA demetilaz, NADH sitokrom bs redüktaz enzim aktiviteleri üzerindeki etkisi araştırıldı. 30 mg /kg thiopental'in intraperitoneal uygulamasını takiben 1, 30 ve 60 dakika sonunda deserebre edilen üç farklı grupta, enzim aktiviteleri çalışıldı. Kontrol ve deney gruplarından elde edilen değerler Kruskal Wallis varyans analizi ve Mann- Whitney U testiyle istatistiksel olarak karşılaştırıldı. Bu sonuçlara göre; karaciğer NADPH sitokrom P450 redüktaz enzimine kontrol grubuna göre 1. grupta % 28, 2. grupta % 57, 3. grupta % 80 aktivasyon; NADH sitokrom bs redüktaz enziminde kontrol grubuna göre 1. grupta % 29, 2. grupta % 32, 3. grupta % 35 aktivasyon saptanmış, NDMA demetilaz enziminde ise anlamlı bir sonuç saptanamamıştır. Akciğerde çalışılan enzim aktivitelerinde ise hiçbir anlamlı sonuç saptanamamıştır. Ayrıca thiopental derişiminin karaciğerde her üç enzim üzerindeki in vitro etkisi de çalışıldı. Bu sonuçlara göre; thiopental derişimi arttıkça karaciğer mikrozomal NADPH sitokrom P450 redüktaz ve NADH sitokrom bs redüktaz enzim aktivitelerinin buna parelel olarak arttığı, NDMA demetilaz enzim aktivitesinin ise azaldığı saptandı.SUMMARY EFFECTS OF THIOPENTAL ON NADPH CYTOCHROME P450 REDUCTASE, NADH CYTOCHROME b5 REDUCTASE AND N-NITROSODIMETHYLAMINE DEMETHYLASE ENZYME ACTIVITIES IN RAT LIVER AND LUNG MICROSOMES The aim of this study was to evaluate the efficacy of thiopental that is a common anaesthetic on NADPH cytochrome P450 reductase, NADH cytochrome bs reductase and N-nitrosodimethylamine demethylase enzyme activities in rat liver and lung microsomes. After the intraperitonally injections of thiopental (30mg/kg) rats were anesthetized and deserebrated at three different periods (1, 30 and 60 min.). Enzyme activities of control and three different treated groups were studied and the results were compared with Kruskal Wallis and Mann- Whitney U test statistically. According to the this results ; the liver NADPH cytochrome P450 reductase enzyme activity increased in three groups was 28%, 57% and 80% of the control group, respectively. The liver NADH cytochrome bs reductase enzyme activity also increased in three groups was 29%, 32% and 35%) of the control group, respectively. No significant change of liver NDMA demethylase activity was observed in any group. Also no significant changes were observed in the rat lung enzyme activities. The in vitro effects of thiopental on liver enzymes also studied.
