Abstract
Katalazın (E.C. 1.11.1.6) hidrojen peroksidi su ve moleküler oksijene yüksek hızda yıkımı, enzimin gıda, optik ve tekstil gibi endüstriyel alanlarda kullanımına yol açmıştır. T. thermophila göl, tatlı su ve su birikintilerinde yaşayan sili bir protozoandır. T. thermophila katalaz (TtCAT1) geninin dizisi Tetrahymena Genom Veritabanından elde edilmiştir. TtCAT1p amino asit dizisinin diğer katalazlar ile dikey hizalanması korunmuş katalaz aktif bölge motifinin (FDRERIPERVVHAKGAG), katalaz hem ligandı bağlanma bölgesi motifinin (RLSYPDTH) ve peroksizomal lokalizasyon sinyalinin (SKI) varlığını göstermiştir. Katalaz geninin mRNA'sının farklı stresler altında Northern Blot analizi, 23°C ile 30°C'de varlığını, aç hücrelerde ise yokluğunu göstermiştir. E. coli'de TtCAT1p'nin rekombinant protein olarak ifadesi için gerekli 10 farklı nokta mutasyonu PZR yöntemiyle yerleştirilmiş ve DNA dizi analizi ile teyit edilmiştir. Rekombinant proteinin E. coli'de üretilip üretilmediği SDS-PAGE ve Western Blot ile anti-GST monoklonal antikor ile anti-sığır katalaz poliklonal antikorları kullanılarak araştırılmıştır. Glutatyon sefaroz afinite boncukları ile saflaştırılmış rekombinant proteinin DOĞAL-PAGE ile katalaz aktivite testi gerçekleştirilmiş, fakat her iki analiz rekombinant GST-TtCat1p proteinin beklenen büyüklükte üretilemediğini ve üretilen ~60 kDa ve ~28 kDa'luk proteinin ise katalaz aktivitesinin bulunmadığını göstermiştir. Sonuç olarak, katalaz geninin açlık şartlarında mRNA'sının bulunmamasının moleküler kontrol mekanizmasının çalışılması için iyi bir deneysel alt yapı oluşturabileceği ortaya konulmuştur. Biyoteknolojik olarak ise TtCatp'in değerlendirilebilmesi rekombinant protein üretim deneylerinin tekrarlanmasını gerektirmektedir. The high rates of decomposition of hydrogen peroxide to water and molecular oxygen by catalase (E.C. 1.11.1.6) causes it to be used in food processing, optics and textile industries. T. thermophila is a ciliated protozoon that inhabits the lakes, fresh waters and ponds. The complete sequence of T. thermophila catalase (TtCAT1) gene was received from Tetrahymena Genome Database. Multiple sequence alignment of TtCat1p amino acid sequence with other catalyses showed the existence of conserved catalase active site motif (FDRERIPERVVHAKGAG), catalase hem ligand binding domain motif (RLSYPDTH) and peroxisomal localization signal (SKI) motif. The analysis of catalase gene mRNA expression by Northern Blot revealed the existence of TtCAT1 mRNA at 23°C and 30°C grown cells but the absence of its mRNA in starved cells. The necessary ten point mutations for the expression of TtCAT1p as a recombinant protein in E. coli were incorporated by in vitro PCR methods and their incorporation to the coding region was confirmed by DNA sequencing. The situation about the expression of recombinant GST-TtCat1p protein was analyzed with SDS-PAGE and Western blot by using anti-GST monoclonal antibody and anti-bovine catalase polyclonal antibody. The purified recombinant GST-TtCat1p with glutathione sepharose beads analyzed with Native-PAGE catalase gel assay. Both methods showed that the expected size recombinant protein was absent but the two truncated forms of ~60 kDa and ~28 kDa proteins was expressed without any catalase activity. In conclusion, the absence of catalase gene mRNA in starved cells suggests that it could be a good assay system to study the molecular control mechanisms of the cat mRNA expression. In term of Biotechnology, the expression of recombinant TtCat1p ought to be repeated to get a fully expressed TtCat1p to study its activity.
