Abstract
Nitrik oksit biyolojik sistemlerdeki birçok fiziksel ve patolojik süreçlerde ve hücrelerde üretilen küçük reaktif oksijen türlerinde yer alan en önemli biyokimyasal parametrelerden biridir. Bu nedenle nitrik oksit için gerçek zamanlı sensör geliştirme çalışmaları biyomedikal araştırmalar ve tanılar için büyük dikkat çekmektedir. Bu tez büyük ölçüde nitrik oksit tayini ve ölçümü çalışmalarında uygulanabilecek optik kimyasal sensör geliştirilmesi üzerinedir. Bu amaçla NO duyarlı moleküller piren, tris(2,2'-bipiridil)diklororutenyum(II) hegzahidrat ve magnezyum fitalosiyanin uygun katkılar ile birlikte polimerik veya camsı matrisler içine gömülmüştür. Matris malzemesi olan polivinil klorür ve sol-jel indikatör boyalarıyla üstün uyumlulukları, suya ve sert koşulları dirençli nitelikleri, indikatör boyalara uygun mikro ortam sağlayarak indikatör boyaları değişimlerden koruma yetenekleri nedeniyle seçilmişlerdir. Bunun yanı sıra, nano boyutta gümüş gibi iletken metalik türlerin varlığı floroforun yayım spektral özellikleri üzerinde etkili olabilir. Bu çalışmada, biz gümüş nano-partikülleri iyonik sıvı ile birlikte polimerik nitrik oksit duyarlı ince filmler ve elektro eğrilmiş fiberlerin üretimi için katkı olarak kullandık. Potansiyel algılayıcı ajanların performansları durgun hal floresans ve zaman çözümlemeli floresans ölçümleri ile test edilmiştir. Bunların arasında piren boyası radikalik nitrik oksite en yüksek yanıtı sergilemiştir. Duyarlılık açısından pireni fitalosiyanin boyası takip etmiştir. Şimdiye kadar piren ve tris(2,2'-bipiridil)diklororutenyum(II) hegzahidrat nitrik oksit algılayıcı çalışmalar için misellerde veya solüsyonlarda test edilmiştir. Bildiğimiz kadarıyla nitrik oksit algılayıcı amaçlarla piren, tris(2,2'-bipiridil)diklororutenyum(II) hegzahidrat ve magnezyum fitalosiyanin kullanan katı hal çalışması bulunmamaktadır. Nitric oxide is one of the most important biochemical parameters taking place in numerous physical and pathological processes in biological systems. Therefore real-time sensor development studies for nitric oxide attract great attention for biomedical research and diagnosis. This thesis is largely concerned with the development of optical chemical sensors for nitric oxide that can be applied to detection and quantification studies. For this purpose nitric oxide sensitive molecules Pyrene, tris(2,2'-bipyrdyl)dichlororuthenium(II) hexahydrate and magnesium phthalocyanine were embedded in polymeric or glassy matrices along with appropriate additives. Matrix materials of polyvinyl chloride and sol-gel were chosen due to excellent compatibility with the indicator dyes, resistant characteristics to water and harsh conditions providing an appropriate micro environment for them. On the other hand, it is known that presence of the conducting metallic species like nano-sized silver may be effective on emissive spectral properties of nearby fluorophores. In this work, we utilized the silver nanoparticles along with ionic liquid as additives for fabrication of polymeric nitric oxide sensitive thin films and electrospun fibers. The performances of the potential sensing agents were tested by steady state fluorescence and decay time measurements. Among them the pyrene dye exhibited the highest response for radicalic nitric oxide. The phthalocyanine dye followed the pyrene in terms of sensitivity. Up to now the pyrene and tris (2,2'-bipyrdyl)dichloro ruthenium(II) hexahydrate have been tested in micelles or solutions for nitric oxide sensing studies. To the best of our knowledge there is no reported solid state study utilizing the pyrene, tris(2,2'-bipyrdyl)dichlororuthenium(II) hexahydrate and magnesium phthalocyanine for nitric oxide sensing purposes.
