Kolorektal kanserli hastalarda hox 2C ve hox 4B homeobox genlerin transkripsiyonlarının RT-PCR tekniği ile araştırılması

Abstract
Homeobox genleri (Hox genleri), insanda 4 farklı kromozom üzerinde lokalize olmuş kırk kadar genden meydana gelmiştir. Homeobox genlerin herbirinde ortak 61 amino asidi şifreleyen 183 nükleotid zincirine sahip Homeodomain diye adlandırılan bir bölge bulunmaktadır. Homebox genler için ilk çalışmalar embriyo safhasında yapılmıştır ve embriyonik gelişimin tamamlanmasında anahtar bir rol oynadıkları ifade edilmiştir. Daha sonra lenf sistemi, böbrek, pankreas, prostat, deri ve kemik iliği tümörlerinde Homeobox genlerinden bir kısmının expresyon veya overexpresyon yaptığı tespit edilmiştir. Kolorektal kanserlerinde de Homeobox genlerinin varlığı hakkında bazı çalışmalar yapılmış fakat kullanılan metotların hassasiyetlerinin az olması nedeniyle kesin sonuçlara gidilememiştir. Bu çalışmada Hox 2C ve Hox 4B genlerinden çok az miktarda transkripte edilen mRNA'lardan çıkılarak RT-PCR tekniği ile bu mRNA'larının milyonlarca defa çoğaltılması hedeflenerek bu genlerin transkripsiyonları hakkında kesin bilgi sahibi olunması amaçlanmıştır. Kolorektal kanserli 10- hastadan kolektomi esnasında tümörlü ve normal doku örnekleri sıvı azot içerisine alınarak dondurulmuştur. Tümörlü ve ayrıca normal dokulardan total RNA'lar guanidyum tiyosiyanat yöntemi ile saflaştırılmıştır. Oligo (dT) primeri ile LcDNA'ların sentezinden sonra Hox 2C ve Hox 4B'nin spesifik primerleri kullanılarak sırasıyla 445 ve 301 bp'lik PCR ürünleri çoğaltılmaya çalışılmıştır. PCR ürünleri % 2'lik agaroz jelde yürütülmüş ve etidyum bromid ile incelenmiştir. On hastanın sadece dördünün normal dokularında Hox 2C geninin transkripsiyonu belirlenmiş, kanserli dokularında ise daha kalın bantlar elde edilmiş ve bu durum semikantitatif manada overtranskripsiyon olarak yorumlanmıştır. Bir hastanın normal dokusunda Hox 2C ile ilgili bir bant gözlenmezken tümörlü dokusunda ise hafif bir bant görülmüştür. Tüm hastaların normal ve kanserli dokularında Hox 4B geninin transkripsiyonuna rastlanmamıştır. Kolorektal kanser vakalarında diğer genlere ilave olarak Homeobox genlerinin transkripsiyonları da dikkate alınmalıdır. Homeobox genes have been composed of 40 genes localized on different 4 chrosomes (2, 7, 12, 17). In each homeobox gene, there is a common region called Homeodomain, which has 183 bp coding 61 amino acids. Early studies about homeobox genes were performed in embryonic stage and it was reported that these genes play a key role in embryonic development. Recently, it was determined that some of homeobox genes have expression or overexpression in tumors of lymphoid, kidney, pancreas, prostate, skin, and bone marrow. Some studies were also done for identification of homeobox genes in colorectal cancer. However, correct results could not be found for these genes, since the used methods have less sensitivity. In the present study, it was aimed to obtain correct results about transcription of these genes by using RT-PCR assay, which amplifies as millions as in little quantitive transcribed mRNAs of Hox 2C and Hox 4B. Nontumorous and tumor samples were obtained during colectomy from 10 patients with colorectal cancer. All samples were frozen in liquid nitrogen. Total RNAs were purified by the guanidinum thiocynate tecnique from nontumorous and tumor samples. Amplification of PCR products of 445 and 301 bp of Hox 2C and Hox 4B homeobox genes, respectively, were studied by using specific primers of these genes after 18t cDNAs were produced by using oligo (dT) primer. The PCR products were run on 2 % agaros gel and were investigated by staining of ethidium bromide. Transcription of Hox 2C gene in nontumorous samples was determined in only four of ten patients, and overtranscription of Hox 2C gene was also observed semiquantitatively in tumor samples of the same four patients. Transcription of Hox 2C gene in one patient was determined in only tumor sample. Transcription of Hox 4B gene was not seen in nontumorous and tumor samples of all patients. In addition to the other genes, transcriptions of Homeobox genes should be taken into consideration for colorectal cancer research.
