Abstract
EX ÖZET Bu çalışmada, Usnea longissima Ach. liken türünden izole edilen difraktaik asit (DA)'in ratlarda indometazin (İND) ile oluşturulan ülser modelindeki koruyucu etkisi (in vivo) araştırıldı. Her bir deney grubu 6'şar rattan oluşturuldu. 25, 50, 100 ve 200 mg/kg dozlarda DA, 50 mg/kg dozda ranitidin (RAN, pozitif kontrol) ve 25 mg/kg dozda İND (negatif kontrol) oral yolla verildi. DA'yı süspansiyon haline getirmek için kullanılan (%1'lik CMC) çözeltisi İND ile birlikte ayrı bir grup olarak deneye alındı. DA'nm olası etkilerini gözlemek için bir deney grubu da İND uygulamaksızın yalnızca 100 mg/kg dozda DA verildi. Çalışmalarda kontrol grubu olarak kullanılmak üzere bir gruba da musluk suyu verildi. İND ile muamele edilen grupta meydana gelen ülserin, uygulanan DA'nın her dört dozu vasıtasıyla önemli oranda (p<0.05) azaltıldığı belirlendi. Diğer yandan, antioksidant savunma sistemlerinin ülser gelişimindeki rolünün belirlenmesi amacıyla, rat mide dokularında süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon peroksidaz (GPx) gibi antioksidan enzimlerinin aktiviteleri ile total glutatyon (GSH) ve lipit peroksidasyon (LPO) miktarları belirlendi. Bulgular, kontrol grupları ile karşılaştırılarak değerlendirildi. İND uygulanan dokularda GSH miktaı ile SOD ve GPx enzim aktiviteleri azalırken, CAT aktivitesiyle LPO miktarının arttığı tespit edildi. Elde edilen sonuçlar, gastrik hasar oluşumunda serbest radikallerin üretildiğini ve İND'nin antioksidan savunma sistemini olumsuz etkileyerek ülser oluşumuna katkı sağladığını göstermektedir. Usnea longissima Ach' dm saflaştırılan DA ve RAN ile muamele edilmiş dokularda, bu maddelerin İND'nin aksine antioksidan savunma sistemini olumlu yönde etkilediğini ve gastrik mukozada üretilen reaktif oksijen (ROS) radikallerinin ülser oluşumundaki olumsuz etkilerinin azaltıldığı bulunmuştur. X SUMMARY An investigation on the protective effects of diffractaic acid isolated from Usnea longissima Ach. on the indomethacine-induced ulcer model in rats, and its in vivo antioxidant properties. In this study, the gastro-protective effect of diffractaic acid (DA) isolated from a lichen species, Usnea longissima Ach. in indomethacin induced ulcer model in rats was investigated, in vivo. Each of the experimental groups consisted from 6 rats. DA at 25, 50, 100 and 200 mg/kg doses, ranitidine at 50 mg/kg dose (positive control) and indomethacin at 25 mg/kg dose (negative control) administrated per orally. %1 CMC solution, used to prepare homogeneous suspension of DA, with indomethacin is taken under control as another group. 100 mg/kg of DA is administrated to healthy rats without indomethacin to understand the effects of DA. It is determined that ulcers occurred in indomethacin administrated tissues were decreased significantly by all doses of DA (p<0.05). In order to discuss the role of antioxidant defence systems on ulcer progress, activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), and total glutathione (GSH) and lipid peroxidation (LPO) amounts were determined in rat stomach tissues. Results appreciated by comparing with control groups. In indomethacin administrated tissues, increased CAT activity and LPO amount in contrast to decreased activities of SOD and GPx and GSH amount were fixed. These results suggested that free radicals are produced in the gastric mucosal damage and indomethacin also effects negatively to the antioxidant defence systems that contribute ulcer formation. It is appeared that, in DA, isolated from Usnea longissima, and ranitidine administrated- tissues, antioxidant defence system was affected affirmative in contrast to indomethacine-administrated tissues, and negative effects of reactive oxygen (ROS) radicals, produced in gastric mucosa were reduced.
