Abstract
Amaç: Bu çalışmanın amacı, ISO toksisistesine bağlı ölen ratlarda postmortem beyin ve böbrek dokusunda UTR ve ET-1 seviyesinde meydana gelen değişimleri araştırmaktır.Materyal ve Metot: Çalışmada ratlar kontrol ve ISO toksisite grubu olarak ayrıldı. ISO toksisitesi, tek lethal doz (850 mg./kg.) ISO intraperitoneal uygulanarak oluşturuldu. Grupların postmortem olarak incelenen saatlerde (0, 1, 3, 6, 12, 24, 48 ve 72.) beyin ve böbrek dokusunda UTR ve ET-1 seviyeleri, plazmada UT, BUN ve kreatinin seviyeleri incelendi. Beyin ve böbrek dokularının histopatolojik görünümleri değerlendirildi.Bulgular: Çalışmamızda ISO toksisite grubunda; plazma UT seviyesinde ve plazma kreatinin düzeyinde artış, histopatolojik olarak beyinde iskemik değişiklikler izlendi. Aynı grupta UTR ekspresyon seviyesinde beyinde 1. saatten itibaren, böbreklerde 3. saatten sonra düşme saptandı. ET-1 seviyesinde beyinde 48. saate kadar devam eden artma, böbrekte 0. saatten itibaren düşme tespit edildi.Sonuç: URT ve ET-1 seviyelerinde gözlenen değişiklikler, postmortem incelemelerin yapıldığı adli tıp alanında klinik uygulamalar açısından değerli olabilir. Kardiyak şoka bağlı ani ölümlerin postmortem değerlendirilmesinde kullanlabilir. Beyin ve böbrek dokusunda UTR ekspresyon ve ET-1 seviyelerindeki belirlenen değişikliklere göre, UT ya da ET-1 agonist veya antagonistlerinin beyin ve böbrekte iskemik değişikleri önlemede faydalı olabileceği ile ilgili araştırmalar yapılabilir. Aim: The aim of this study was investigating changes in postmortem UTR and ET 1 levels of brain and kidney tissues in rats died because of ISO toxicity.Material and Method: In the present study, the rats were divided into two groups as control and ISO toxicity groups. ISO toxicity was created by applying a single lethal dose of (850 mg/kg) ISO intraperitoneal. UTR and ET-1 levels in brain and kidney and UT, BUN and creatinine levels in plasma were investigated at postmortem hours (0, 1, 3, 6, 12, 24, 48 and 72nd). Histopathologic appearance of the brain and kidney tissues were evaluated.Results: In our study, in the ISO toxicity group, an increase was found in plasma UT and plasma creatinine levels. It was observed ischemic changes as histopathological in the brain tissue. In the same group, a reduction was observed in UTR expression level after the 1st hour in the brain and after the 3rd hour in kidneys. On the other hand, ET-1 level was increased in the brain until the 48th hour, whereas it was observed to be reduced in the kidney from the beginning.Conclusions: Observed changes in UTR and ET-1 expression level might be valuable in clinical practices in the field of forensic medicine where postmortem examinations are conducted. They can be used in the postmortem evaluation of sudden death caused by cardiac shock. Further studies may be conducted to show that UT or ET-1 agonist or antagonists may be useful in preventing ischemic damage in the brain and kidneys according to the changes determined in UTR expression and ET-1 levels in the brain and kidney tissue.
