Abstract
Amaç: Bakteriyel enfeksiyonların neden olduğu sistemik inflamatuar bir yanıt olan sepsisin patogenezinde endotel hücre hasarının anahtar rol oynadığı in-vivo ve in-vitro çalışmalarla kanıtlanmıştır. Bununla birlikte, yüksek antiinflamatuar etkiye sahip olan Myrtus communis L. (MC) bitkisinin meyvelerine ait uçucu yağ ve bileşenlerinin, antiinflamatuar mediatörler üzerine de etkili olduğu da belirlenmiştir. Bu sebeple bu çalışmada, MC bitkisinin meyvelerine ait uçucu yağ ve bileşenlerinin; sepsisin neden olduğu endotel hücre hasarına olan etkileri in vitro sepsis modelinde moleküler ve hücresel açıdan incelenmiştir.Materyal ve Metot: Çalışmamızda, insan göbek kordonu veni endotel hücre hattı (HUVEC) kullanıldı. Lipopolisakkarit (LPS) ile indüklenen sepsis modelinde, gruplar kontrol, LPS, MC, MC+LPS, 1.8 cineole, 1.8 cineole+LPS, α-pinene, α-pinene+LPS, α-terpineole, α-terpineole+LPS olacak şekilde belirlendi. Hücre proliferasyonları xCELLigence® sistemi ile analiz edildi. Ayrıca, tümör nekrozis faktörü (TNF-α), interlökin-1β (IL-1β), interlökin-6 (IL-6) ve eNOS (endotelyal nitrik oksit) sitokinlerinin mRNA expresyonları kantitatif polimeraz zincir reaksiyon (qPCR) analizi kullanılarak tespit edildi.Bulgular: Sonuç olarak, MC, terpineole, 1.8 cineole ve α-pinene bileşiklerinin HUVEC hücre hatlarına belirli dozlarda uygulanması kontrol grubuna göre hücresel indekste bir değişikliğe neden olmamıştır. HUVEC hücre hatlarına LPS uygulaması ise zamana bağlı olarak hücresel indekste anlamlı derecede azalmaya neden olmuştur. MC+LPS ve α-terpineole+LPS gruplarının LPS kontrol grubuna göre azalmış hücre indeksini düzeltememiş; 1.8 cineole+LPS ve α-pinene+LPS bileşiklerinin ise LPS kontrol grubuna göre azalmış hücre indeksini anlamlı derecede artırdığını kontrole yaklaştırdığı tespit edilmiştir. qPCR analiz sonuçlarına göre; LPS ile indüklenmiş hücresel hasarda MC uçucu yağı ve α-terpineole bileşiklerinin uygulanması artmış olan TNF-α, IL-1β, IL-6 ve eNOS mRNA ekspresyonlarını anlamlı derecede düşüremezken; 1.8 cineole ve α-pinene bileşiklerinin uygulaması ise LPS ile indüklenmiş TNF-α, IL-1β, IL-6 ve eNOS mRNA ekspresyonlarını anlamlı derecede düşürdüğü gözlemlenmiştir. Sonuç: MC bitkisinin meyvelerinden elde edilen uçucu yağın ve MC bitkisinin meyvelerine ait bileşiklerden α-terpineole bileşiğinin LPS ile oluşturulmuş endotel hücre hasarına bağlı gelişen azalmış hücre indeksine ve artmış sitokin cevabına etkilerinin bulunmadığı gösterilmiştir. Bununla beraber MC bitkisinin meyvelerine ait diğer major bileşenlerden olan 1.8 cineol ve α-pinene moleküllerinin LPS ile oluşturulmuş HUVEC hücre hasarını hücresel indeks ve biyomoleküler seviyede (TNF-α, IL-1β, IL-6 ve eNOS) düzelttiği gösterilmiştir. Aim: In-vivo and in-vitro studies have demonstrated that endothelial cell damage plays a key role in the pathogenesis of sepsis, a systemic inflammatory response caused by bacterial infections. However, it has also been determined that essential oils and components of the fruit of the Myrtus communis L. (MC) plant, which has a high antiinflammatory effect, are also effective on antiinflammatory mediators. For this reason, this study also showed that the essential oils and components of the fruits of the MC plant, The effects of sepsis on endothelial cell damage were investigated in terms of molecular and cellular in vitro sepsis model.Material and Method: In our study, human umbilical cord vein endothelial cell line (HUVEC) was used. In the sepsis model induced by lipopolysaccharide (LPS), the groups were identified as LPS, MC, MC+LPS, 1.8 cineole, 1.8 cineole+LPS, α-pinene, α-pinene +LPS, α-terpineole, α-terpineole+LPS. Cell proliferations were analyzed by the xCELLigence® system. In addition, mRNA expressions of tumor necrosis factor (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6) and eNOS (endothelial nitric oxide) cytokines were determined using quantitative polymerase chain reaction (qPCR) analysis.Result: As a result, administration of MC, terpineole, 1.8 cineole and α-pinene compounds at specific doses to HUVEC cell lines did not cause a cellular index change compared to the control group. The application of LPS to HUVEC cell lines led to a significant decrease in cellular index, depending on the time. MC + LPS and α-terpineole + LPS groups did not resolve the decreased cell index compared to the LPS control group; It has been found that 1.8 cineole + LPS and α-pinene + LPS compounds increase the decreased cell index significantly compared to the LPS control group. According to qPCR analysis results; The application of MC volatile oil and α-terpineole compounds to LPS-induced cellular damage significantly reduced TNF-α, IL-1β, IL-6 and eNOS mRNA expression levels; The application of 1.8 cineole and α-pinene compounds significantly reduced LPS-induced TNF-α, IL-1β, IL-6 and eNOS mRNA expression.Conclusion: It has been shown that the α-terpineole compound from the volatile oils obtained from the fruits of MC plant and the fruits of the MC plant has no effect on the decreased cell index and increased cytokine response due to LPS-induced endothelial cell damage. However, it has been shown that 1.8 cineol and α-pinene molecules, one of the major components of MC fruits fruiting, correct LPS-induced HUVEC cell damage at the cellular index and biomolecular level (TNF-α, IL-1β, IL-6 and eNOS).Key Words: HUVEC, LPS, Myrtus communis L., sepsis, 1.8 cineole, α-pinene, α-terpineole
