Abstract
Amaç: Etofenamat etken maddesinin farmasötik preparatlarda miktar tayinine yönelik UV-Görünür Bölge Absorpsiyon Spektroskopi, Birinci Derece Türev Spektroskopi ve HPLC yöntemlerini geliştirmek, valide (geçerlilik testlerini) etmek ve uygulanabilir olduğunu göstermek için etofenamate etken maddesini içeren farmasötik preparatlarda etofenamate miktar tayininin yapılması amaçlanmaktadır.Materyal ve Metot: UV-Görünür Bölge Absorpsiyon Spektroskopi ve Birinci Derece Türev Spektroskopi yöntemlerinde, metanolde etofenamatın spektrumları alındı ve en iyi absorbans değerleri sırasıyla 286 nm ve 298 nm dalga boyları belirlendi ve çalışmada kullanıldı. HPLC yönteminde ise metanol:asetonitril:su (45:35:25, h/h/h) hareketli fazı, C18 ters faz kolonu, 1 mL/dk akış hızı, 10 μL enjeksiyon hacmi, iç standart (IS) cilostazol ve 286 nm dalga boyundan oluşan çalışma parametreleri kullanıldı. Her üç yöntemin çalışma parametreleri belirlendikten sonra doğrusallık, analitik geri kazanım, güniçi ve günler arası doğruluk ve kesinlik gibi parametreleri içeren geçerlilik testleri yapıldı.Bulgular: 0.5-45 μg/mL derişim aralığında HPLC yönteminin, 1-50 μg/mL derişim aralığında UV-Görünür Bölge Absorpsiyon Spektroskopi ve Birinci Derece Türev Spektroskopi yöntemlerinin doğrusal olduğu tespit edildi. Güniçi ve günler arası kesinlik ve doğruluk değerlerinin her üç yöntem için sırasıyla %7.92 ve %3.11'den daha iyi, analitik geri kazanım değeri ise her üç yöntem için ortalama olarak % 98.50 olarak belirlendi.Sonuç: Etofenamate için geliştirilen ve validasyonu yapılan HPLC, UV-Görünür Bölge Absorpsiyon Spektroskopi ve Birinci Derece Türev Spektroskopi yöntemlerinin hassas, duyarlı, seçici, doğru ve kesin olduğu validasyon çalışmalarıyla gösterildiğinden dolayı yöntemlerin farmasötik preparatlarda kalite kontrol amaçlı çalışmalarda başarıyla uygulanabilir olduğu sonucuna varıldı.Anahtar Kelimeler: Birinci Derece Türev Spektroskopi Yöntemi, Etofenamate, Farmasötik Preparat , HPLC, UV-Görünür Bölge Absorpsiyon Spektroskopi Yöntemi. Aim: It is aimed to determine the quantity of etofenamate in pharmaceutical formulations involving etofenamate agent in order to develop UV-Visible Absorption Spectroscopy, First-Order Derivative Spectroscopy and HPLC methods, validate (validity test) and show the applicability of methods developed for the determination of the quantity etofenamate agent in pharmaceutical formulations.Material and Method: In the UV-Visible Absorption Spectroscopy and First-Order Derivative Spectroscopy methods, the spectrum of etofenamate was taken in methanol and optimum absorbance values were obtained in the wavelengths of 286nm and 298nm . In HPLC method, mobile phase (methanol:acetonitrile:water (45:35:25, v/v/v)), reverse phase column (C18), flow rate (1 mL/min), injection volume (10 μL), cilostazol as internal standard (IS) and wavelength (286 nm) were used as working parameters.After the determination of the working parameters of all three methods, validity tests including linearity, analytical recovery, intraday and inter-day accuracy and precision were performed.Results: It was found that HPLC and UV-Visible Absorption Spectroscopy and First-Order Derivative Spectroscopy methods were linear in the concentration ranges of 0.5-45 μg/mL and 1-50 μg/mL respectively. The intra- and inter-day accuracy and precision rates for both methods were beteer than %7.92 and %3.11 respectively and the analytic recovery values for both methods were found to be 98.5% on the average.Conclusion: It was concluded that the developed and validated HPLC and UV-Visible Absorption Spectroscopy methods for etofenamate could successfully be applied on pharmaceutical preparation since HPLC, UV-Visible Absorption Spectroscopy and First-Order Derivative Spectroscopy methods developed and validated for etofenamate were shown to be sensitive, selective, accurate and precise.Keywords: First Order Derivate Spectroscopy method, Etofenamate, Pharmaceutical Preparation, HPLC, UV-Visible Absorption Spectroscopy method.
