Erzurum`da çiğ olarak yenilen çeşitli sebzeler ile içme sularından izole edilen Escherichia coli suşlarının değişik özellikleri üzerinde araştırmalar

Abstract
Erzurum piyasasında satılan ve çiğ' olarak yenilebilen may-danoz, marulÎ, taze soğan, nane, lahana, karnıbahar, salatalık,havuç, dere otu, domates gibi sebze örneklerinden ve şehrin içme suyu ör-neklerinden izole edilen E.coli suşlarınm, biyokimyasal özellikle` ri, hemoliz aktivitesi, koüsinojenite ve lizojenite özellikleri çe¬şitli antibiyotiklere duyarlılık durumu ve bulaşıcı tip plazmid (R-faktör) taşıma özellikleri üzerinde araştırmalar yapılmıştır.Çalışmada 300 adet 11 cins ve türe ait sebze örneği yıkan¬madan ve yıkandıktan sonra incelenmiştir. Yıkanmamış sebze örnekle™ rinden 216 smda E.coli'ye rastlanılmış ve her birinden 3 suş olmak üzere 648 adet suş izole edilmiştir. Aynı örnekler normal bir yıka¬mayla yıkandıktan sonra, tekrar muayeneye tabi tutulmuş ve 140 numu¬nede gene E,coli7ye rastlanılmış her birinden 3 suş olmak üzere 420 suş ayrılmıştır. Şehrin içme sularından alman 300 adet numuneden 160 mda E.coli*ye rastlanılmış ve her birinden 3 suş olmak üzere 480 suş izole edilmiştir. Sebze ve su örneklerinden toplam olarak 1548 adet suş üzerinde çalışılmıştır.Yapılan bu çalışmalardan elde edilen bulgular aşağıda özet-lenmiştir.1« İncelenen Bmcoii suşlarınm biyokimyasal özellikleri ge-nellikle literatürde verilen standartlarına uygun bulunmuştur. Suş- ların hiç birisinde hemoliz aktivitesine rastlanılmamıştır.2- Yıkanmış ve yıkanmamış sebzelerden izole edilen E%coli suçlarında kolisin özelliğine rastlanılmamasma karşılık, içme su¬lar mdak il erde yüzde 3 2.29 oranında koüsinojenite özelliğine rast-* lanılmıştır.4-Suşiarın hepsi nalidiksik aside duyarlı, eritromisine dirençli bulunmuştur. En yüksek duyarlılık gentamisine karşı olmuş, buna karşılık kloramfenikol ve ampisiline karşı yüksek oranda direnç bulunmuştur. Suşların diğer antibiyotiklere karşı dirençlilikleri farklı olmuştur.5-Yıkanmış, yıkanmamış sebzelerden ve içme sularından izo¬le edilen E + coli suşlarmın antibiyotiklere çoklu direnç gösteren¬ler sırasıyla yüzde 30,00, 29.17 ve 47.50 oranlarında direnç1iliğin hepsini veya bir kısmını transfer etmiş oldukları saptanmıştır,6-Yıkanmamış sebze örneklerinde yüzde 72,00, yıkanmışlar¬da ise yüzde 46.67 arasında E.coli'ye rastlanılmıştır.Bulguların literatür bilgisi ile tartışılması yapılmış/ çiğ yenilen sebze hijyeni ile ilgili Önerilerde bulunulmuştur. Research Works On The Certain Characteristics of E.coli Isolated From The Vegetables Consumed Without Cooking And From The Drinking Water In Erzurum.Research works were carried out on the biochemical charac-teristics f hemolytic activity, colicinogenicitv and lysogenicity properties, sensitivity to the various antibiotics and whether carr¬ying contagious plasmid (R-factor) or not of the E+coli strains iso¬lated from the uncooked consumed vegetables sold in Erzurum markets such asparsley, lettuce, green onions, pepermint, cabbage, cauli¬flower, cucumber, carrot, dill and tomato and from the drinking wa¬ter samples of this same city.In the work, total of 300 vegetable samples belonging to the 11 different variety and species were examined before washing and after the washing* In the 216 samples of the unwashed vegetab¬les E.coli were found and by determining 3 different strains in each vegetable, total of 648 E.coli strains were isolated. The.se same samples were again examined after a normal washing. This time, in only 14 0 samples E.coli were found and with 3 different strains in each, so, total of 420 strains were isolated from the drinking water supplies of the city 300 samples were collected and in the 160 of these water samples E.coli were found and with again 3 different strains in each sample, total of 480 strains were isolated. Thus, it was worked on the total of 1548 strains drived from the vegetab¬le and water samples.The obtained results from these works were summarized below,1-The biochemical characteristics of the examined B.coli strains were generally found to be fitting to the standards of the literatures. In none of the strains hemolytic activity was determi¬ned .2-Although, the colicinogenicity properties were not de~ termined in the Emcoli strains isolated from the washed and unwas¬hed vegetables, the strains of the drinking water samples showed a 32.29 % colicinogenicity characteristic,3~ It was determined that E.coli strains, from the unwas¬hed and washed vegetables and from the drinking water samples, had respectively, 1.70, 1.67 and 21.88 percent lysogenicity characteris¬tics .4-It was found that all of the strains were sensitive to the nalidixic acid and resistant to the erythromycin. The highest sensitivity was determined against to gentamycin, but contrary, it was found a high resistant against to chloramphenicol and ampicilin. The resistance of the strains to the other antibiotics were found to be varied.5-It was also determined that the isolated E.coli strains, from the washed, unwashed vegetables and from the drinking water, which showing a polyway resistance to the antibiotics, transferred their resistivity in whole or in part at the 30.00, 29.17 and 47.50 percent respectively*6` E.coli was found to be present in the 72.00 % of the unwashed and 46,67 % of the washed vegetable samples.Obtain results were discussed with the literature datas andI'V' >■ ■ •*some proposals were given for the hygiene of the vegetables consu¬med without cooking.
