Meristem üç kültürü ile Granola pasinler 92 ve Caspar patates (Solanum Tuberosum L.) çeşitlerinden PVX, PVY ve PLVR virüslerinden arındırılmış mikro yumru elde edilmesi

Abstract
ÖZET Bu araştırmada meristem uç kültürü tekniği kullanılarak patates X virüsü (PVX), patates Y virüsü (PVY) ve patates yaprak kıvırcıklık virüsü (PLRV) ile enfekteli Granola, Pasinler 92 ve Caspar patates çeşitlerinden bu virüslerden arındırılmış mikro yumru elde etmek amaçlanmıştır. Meristem gelişimi ve in vitro çoğaltım için en uygun Benziladenin (B A) ve Gibberallik asit (GA3) kombinasyon ve konsantrasyonun belirlenmesinde Murashige ve Skoog (MS) temel besi ortamına farklı oranlarda BA ve GA3 ilave edilmiştir. In vitro şarlarda mikro yumru üretiminde ise BA, Chlorocholine chloride (CCC) ve ışığın etkileri incelenmiş ve çalışmaların her aşamasında bitkicikler ELISA ile testlenmişlerdir. Çalışmalarda, Granola, Pasinler 92 ve Caspar çeşitlerinde meristemlerin sırasıyla 0.50/0.50, 0.50/0.25 ve 0.50/0.50 mg/1 BA/GA3 içeren besi ortamında daha iyi geliştikleri belirlenmiştir. Diğer taraftan, bu çeşitlerin sırasıyla 0.00/0.50, 0.00/0.50 ve 0.00/0.25 mg/1 BA/GA3 uygulamalarında daha fazla nod oluşturdukları ve daha iyi geliştikleri tespit edilmiştir. Mikro yumru üretimi çalışmalarında çeşitlerden en fazla sayıda yumru her üç çeşit içinde 5.0/500 mg/1 BA/CCC içeren uygulamadan elde edilmiştir. Çeşitlerden virüslerin eleminasyonuna yönelik çalışmalarda sadece meristem uç kültürü tekniği kullanılmış ve meristemler maksimum 0.3 mm uzunluğunda kesilmişlerdir. Sonuç olarak, Granola, Pasinler 92 ve Caspar çeşitlerinden PVX sırasıyla % 25.0, % 16.0, % 28.5; PVY % 40.0, % 41.6, % 33.3; PLRV ise % 60.0, % 46.1, % 50.0 oranlarında elemine edilmiştir. SUMMARY Experiments were conducted in order to obtain virus-free microtubers from Granola, Pasinler 92 and Caspar {Solanımı tuberosum L.) potato cultivars infected with potato virus X (PVX), potato virus Y (PVY) and potato leaf roll virus (PLRV) by using meristem tips culture. In order to determine the best medium for development of meristem and in vitro clonal propagation of the potato varieties, Murashige ve Skoog (MS) medium containing different combination and concentration of Benziyladenine (BA) and Gibberellic acid (GA3) were tested. For the production of microtubers in in vitro conditions, the effects of BA, Chlorocholine chloride (CCC) and light were studied. All of the plant materials that produced in vitro were checked by enzyme-linked immunosorbent assay (ELISA) to determine whether or not virus-free. The results showed that meristem tips from Granola, Pasinler 92 and Caspar cultivars were grew better on the 0.50/0.50, 0.50/0.25, and 0.50/0.50 mg/1 BA/GA3 treatments, respectively. On the other hand, these cultivars had more nodes and better growth in 0.00/0.50, 0.00/0.25, and 0.00/0.50 mg/1 BA/GA3 treatments respectively. There were no differences between cultivars in terms of microtuber production. All potato varieties tested in this study produced more tubers in 5.0/500 mg/1 BA/CCC treatments under continuos dark period. For the elimination of viruses from potato varieties, only meristem tip culture technique was used, and meristem tips of maximum 0.3 mm length were excised. In conclusion, the elemination percentage of Granola, Pasinler 92 and Caspar cultivars; PVX was 25.0 %, 16 0 %, 28.5 %; PVY was 40.0 %, 41.6 %, 33.3 % and PLRV was 60.0 %, 46.1 % and % 50.0 % respectively.
