Dr. Filiz ERTUNÇ Bu çalışma Malatya ilinde sert çekirdeklilerin özellikle de kayısıların virüs hastalıkları bakımından durumunu araştırmak amacıyla yapılmıştır. Malatya'nın 12 ilçesinde yapılan surveyler sonucu Apple chlorotic leaf spot trichovirus (ACLSV) ve Apple mosaic ilarvirus (ApMV) enfeksiyonlarına rastlanmıştır. Yeşilyurt ilçesi Gündüzbey köyündeki kirazlarda ACLSV; Merkez ilçede bir kayısı ağacında, Battalgazi ilçesi merkezi ve Yazıhan ilçesi Sürür köyündeki eriklerde ApMV, iki şeftali ağacında ise ACLSV + ApMV tespit edilmiştir. Virusların tanısı otsu ve odunsu bitkilere inokulasyon ve ELISA testleri ile yapılmıştır. ELISA testleri Apple chlorotic leaf spot trichovirus (ACLSV), Apple mosaic ilarvirus (ApMV), Arabis mosaic nepovirus (AMV), Cherry leaf roll nepovirus (CLRV), Plum pox potyvirus (PPV), Prune dwarf ilarvirus (PDV), Prunus necrotic ringspot ilarvirus (PNRSV), Raspberry ringspot nepovirus (RRV), Stravvberry latent ringspot nepovirus (SLRSV), Tomato black ring nepovirus (TBRV) ve Tomato ringspot nepovirus (ToRSV) antiserumları kullanılarak yapılmıştır. Yörede ApMV'ye göre daha fazla ağaçta saptandığından dolayı ACLSV saflaştırılmış ve ona karşı 1/256 titrasyonda antiserum elde edilmiştir. Prof. Dr. Filiz ERTUNÇ This study was conducted to search the virus disease status of stone fruit trees, especially apricots, in Malatya Region. As a result of surveys made in 12 districts of Malatya, Apple chlorotic leaf spot trichovirus (ACLSV) and Apple mosaic ilarvirus (ApMV) were found. ACLSV was determined on sweet cherry trees in Gündüzbey village of Yeşilyurt district; ApMV was determined on a apricot tree in Central district, plum trees in Battalgazi district and Sürür village of Yazihan district while ACLSV + ApMV infections were detected on peach trees in Gündüzbey village of Yeşilyurt district. Occurrence of the viruses was proved based on the inoculation tests on woody and herbaceaus indicator plants and ELISA tests. ELISA tests were used for the detection of Apple chlorotic leaf spot trichovirus (ACLSV), Apple mosaic ilarvirus (ApMV), Arabis mosaic nepovirus (AMV), Cherry leaf roll nepovirus (CLRV), Plum pox potyvirus (PPV), Prune dwarf ilarvirus (PDV), Prunus necrotic ringspot ilarvirus (PNRSV), Raspberry ringspot nepovirus (RRV), Strawberry latent ringspot nepovirus (SLRSV), Tomato black ring nepovirus (TBRV) and Tomato ringspot nepovirus (ToRSV). ACLSV was purified, because of the fact that ACLSV infections were detected much more frequently than ApMV. Antiserum produced against ACLSV had a titer of 1/256.
