Abstract
ÖZET Doktora Tezi ENTEROBACTER AEROGENES VE PLAZMİDLERİNDEN 2,3-BÜTANDİOL VE ASETOİN ÜRETİMİNDE pH 6-ASETOLAKTAT OLUŞTURAN ENZİMİN ROLÜ A. Dr. Yakup ARICA Bu çalışmada, bakteriyal hemoglobin geninin (vgb) farklı fragmanlarını taşıyan rekombinant Enterobacter aerogenes (Ea) suşları hazırlandı. Bu rekombinant suşların (EapUC8:15 ve EapUC8:16) metabolik yolda pirüvattan sonra verdikleri katabolik ürün a-asetolaktatın verimi, yaban tip Ea ve vgb geni içermeyen rekombinant E. aerogenes (EapUC8)'inkiler ile karşılaştırıldı. Bakteriyel hemoglobin geni taşıyan rekombinant Ea suşların in a-asetolaktatı, bu geni taşımayan kontrollere göre (Ea ve EapUC8) daha yüksek seviyede ürettikleri anlaşıldı. Bu suşlarda, pH 6 - asetolaktat oluşturan enzim aktivitesinin, 2,3- bütandiol üretimi ile doğrudan bağlantılı olduğu belirlendi. Sonuçlar, seçilen konakçıda (E. aerogenes), aktarılan vgb gen fragmanlarına bağlı olarak 2,3- bütandiol üretiminin kontrol edilebileceğini ve ürün veriminin arttırılabileceğini gösterdi, invert ve elektron mikroskop çalışmaları, vgb geni içeren rekombinant suşların, hücre duvarı kalınlığı ve genel morfoloji bakımından Ea ve EapUC8'e kıyasla önemli farklılıklar içerdiğini ortaya koydu. 1999, 68 sayfa Anahtar Kelimeler: Enterobacter aerogenes, rekombinant E. aerogenes (EapUC8, EapUC8:15, EapUC8:16), 2,3-bütandiol, asetoin, pH 6-asetolaktat oluşturan enzim. Prof. Dr. Yakup ARICA In this study, recombinant Enterobacter aerogenes (Ea) strains carrying different fragments of bacterial hemoglobine gene (vgb) were prepared. A comparison was made with the product a-acetolactate yield of recombinant strains (EapUC8:15 and EapUC8:16) and the wild type Ea as well as the recombinant Ea (EapUC8) that lacks the vgb gene on the metabolic pathway after pyruvate. It was understood that the recombinant strains carrying the vgb gene produced higher levels of a -acetolactate, compared to controls (Ea and EapUC8) that did not carry this gene. Also, the activity of pH 6 - acetolactate forming enzyme was found to be directly related to 2,3-butanedioI production in these strains. The results demonstrated that 2,3-butanediol production could be controlled, and the product yield could be increased by transforming vgb gene fragments, in the choosen host (Ea).
