Abstract
Apodemus mystacinus Balkanlar'dan Orta Asya ve Kafkasya'ya kadar geniş yayılışgöstermektedir. Türkiye'de A. mystacinus'un üç alttürü bulunmaktadır. Daha önceki bir çokçalışma da dahil olmak üzere, A. mystacinus'un Türkiye'deki taksonomik durumu problemlidir.Bu çalışmanın amacı, Türkiye'deki A. mystacinus'un DNA markerlarına dayanarak genetikyapısını araştırmak ve populasyon genetiğine katkıda bulunmaktır. Türkiye'deki 9 lokalitedentoplam 18 birey toplandı. A. mystacinus populasyonlarındaki genetik varyasyonun miktarınıortaya koymak için RAPD-DNA marker sistemi kullanıldı. Çalışılan populasyonlar arasındakigenetik ilişkileri göstermek için Nei'nin genetik mesafe ve genetik benzerlik hesaplamalarıkullanıldı. Hesaplamalar POPGENE ve POPULATION bilgisayar programları ile yapıldı.Çalışılan 60 RAPD markerının 14 tanesi 154 polimorfik RAPD bandı ortaya koymuştur. A.mystacinus populasyonları için hesaplanan genetik farklılık Trabzon populasyonunda H=0,0227 (%P= 4,55) ile Muğla populasyonunda H= 0,2045 (%P= 40,91) aralığındadır. A.mystacinus populasyonlarındaki toplam genetik farklılık HT = 0,3087 olarak hesaplandı.Çalışılan populasyonlar arasındaki genetik farklılığı gösteren GST değeri (0,7438) önemliderecede yüksek çıktı. Genetik mesafe verisiyle oluşturulan dendogram 3 kümelenme gösterdi.A. mystacinus kümesindeki gruplanmanın önceden belirlenen alttür kategorileri ile kısmenuyumlu idi.Anahtar Kelimeler: Kayalık Faresi, Apodemus mystacinus, RAPD-PCR, Türkiye. Apodemus mystacinus is widely distributed from Balkans to Middle East and Caucasus.Three subspecies of A. mystacinus are distributed in Turkey. Despite of several previous studies,the taxonomic status of A. mystacinus in Turkey is problematic.The aim of the present study isto survey genetic structure based on DNA markers and to make contribution to the taxonomicstatus, population genetics of A. mystacinus in Turkey. A total of 18 specimens were collectedfrom 9 locations in Turkey. To explore the extent of genetic variation in A. mystacinuspopulations a Randomly Amplified Polymorphic DNA marker system was used. The estimatesof NEI?s standart genetic identity and standart genetic distance were calculated to show thegenetic relationships between studied populations. All estimations were calculated with thePOPGENE and POPULATION softwares. With the 60 RAPD markers tested, 14 of themyielded 154 polymorphic DNA bands. The estimated genetic diversity for A. mystacinuspopulations was ranged from H= 0.0227 (%P= 4.55) for Trabzon and to 0.2045 (%P=40.91) forMuğla population. The total genetic diversity was calculated as HT= 0.3087 in A. mystacinuspopulations from Turkey. GST value calculated was high (0.7438) indicating that geneticdifferentiation among the studied populations was substantial. Dendogram constructed withgenetic distance data contained 3 clusters. The groupings in the A. mystacinus cluster werepartly consistent with the previously assigned subspecific categories.Key Words: Rocky Mouse, Apodemus mystacinus, RAPD-PCR, Turkey.
