Sıçanlarda streptozotosin diyabet ve eksojen C Vitamini (Askorbik asit) uygulamasına timus lenfositlerinin cevabı

Abstract
Bu çalışmada C Vitamininin (Askorbik Asit), streptozotosin (STZ) ile diyabetik yapılmış sıçanların timus lenfositlerine etkisi incelenmiştir. Çalışmada kullanılan sıçanlar kontrol,diyabet ve C Vitamini uygulanan diyabet gruplarına ayrılmıştır. Diyabet, sitrat tamponunda çözülmüş 45 mg/kg STZ'nin intraperitonal yolla tek doz olarak verilmesiyle oluşturulmuştur. C Vitamini (20 mg/kg), intragastrik gavaj uygulamasıyla 21 gün süre ile verilmiştir. Tüm gruplar, 21. günün sonunda tyopental sodyum anestezisi altında dekapite edilmiştir. Timus dokuları derhal çıkarılmıştır. İlk olarak %2.5'luk gluteraldehit ile sonra %1'lik osmium tetroksit ile tespitleri yapılmış, dehidre edilmiş ve araldit ortamına gömülerek bloklanmıştır. Ultramikrotom ile yarı ince ve ince kesitler alınmıştır. Yarı ince kesitler toluidin mavisi ile boyanarak ışık mikroskobunda; ince kesitler ise uranil asetat ve kurşun sitrat ile boyanarak geçirmeli elektron mikroskobunda incelenmiştir. Her grupta hücresel değişikler ve lenfositlerde mitotik aktiviteye bakılmıştır. Diyabetin mitotik aktiviteyi azalttığı ve mitoz anomalilerine sebep olduğu görülmüştür. C Vitamini uygulanan diyabetik grupta ise kontrol grubununkine yakın mitotik aktivite dikkat çekmiştir. Diyabetik grupta, hem korteks hem de medulla bölgesindeki lenfositlerde ve epitelyal retiküler hücrelerde dejenerasyonlar tespit edilmiştir. C Vitamini uygulanan diyabetik gruptaki hücre hasarının, diyabetik grup ile karşılaştırıldığında, nisbeten daha az olduğu gözlenmiştir. Sonuçlar, diyabetiklere Vitamin C uygulanmasının lenfosit dejenerasyonuna karşı koruyucu etkisi olabileceğini ve mitotik aktiviteyi artıracağını işaret etmektedir.Anahtar kelimeler: STZ diyabet, C vitamini, mitotik aktivite, timik lenfositler In this study, the effects of Vitamin C (Ascorbic Acid) on the thymic lymphocytes of Streptozotocin (STZ) ? induced diabetic rats were studied. The rats were divided into control, diabetic and Vitamin C treated diabetic groups. Diabetes was induced by a single dose of STZ (45 mg/kg) administered intraperitoneally in sodium citrate buffer. Vitamin C (20 mg/kg) was administered intragastrically for 21 days. At the end of 21st day, all animals were decapitated under thiopental sodium anesthesia. The thymus tissues were immediately removed; first fixed in 2.5% glutaraldehyde and then 1% osmium tetroxide; dehydrated; embedded into araldite and blocked. Semithin and thin sections were cut with ultramicrotom. Semithin sections were stained with toluidine blue and examined with light microscopy. Thin sections were stained with uranyl acetate and lead citrate and examined with transmission electron microscopy. Cellular changes and mitotic activity of lymphocytes were evaluated in all groups. It was found that diabetes reduced mitotic activity and caused mitotic anomalies. The mitotic activity in the Vitamin C treated diabetic group was comparable to that of the control group. The cortex and medulla lymphocytes and the epithelial reticular cells were degenerated in the diabetic group. The cell damage was lessened in the Vitamin C treated diabetic group. The results suggest that Vitamin C treatment could protect against lymphocyte degeneration and increase the mitotic activity in the diabetics.Key words: STZ diabetes, Vitamin C, Mitotic activity, Thymic lymphocytes
