Sendromsuz dudak damak yarıklı Türk hastalarda, bazı mutasyon tiplerinin belirlenmesi

Abstract
Dudak ve/veya damak yarıkları konjenital (doğuştan) bozuklukların en yaygın sınıflarından birini temsil etmektedir. Farklı populasyonlarda, coğrafik yerleşim, etnik köken ve sosyoekonomik koşullara bağlı olarak, her 500- 2500 doğumda bir bireye etki ettiği saptanmış olan genel bir sağlık problemidir. Sendromsuz dudak damak yarıklarının oluşumunda genetik faktörlerin rollerine ilişkin çalışmaların çoğu hayvan modellerinde aday genlerin yapıları ve ekspresyon düzeylerinin araştırılması üzerine olmuştur. Bu çalışmada Türk toplumunda, başka populasyonlarda sendromsuz dudak damak yarıklı vakalarda saptanan aday genlerden TGF? ve TGFß3'deki mutasyonların tanımlanması ve bu aday genlerin Türk populasyonundaki sıklığının saptanması amaçlanmıştır. Çalışma için 70 sendromsuz dudak damak yarıklı hasta birey, 85 kontrol birey kullanılmıştır. Sendromsuz dudak damak yarıklı hastalardan elde edilen DNA'larda, TGF? ve TGFß3 genlerindeki mutasyonların araştırılmasında ilgili genlere ait belirli ekzon bölgelerinin PZR'ları yapılmıştır. Ve RFLP işlemlerine geçilmiştir. TGF? geni için elde edilen PZR ürünü HinfI restriksiyon enzimi, TGFß3 geni için elde edilen PZR ürünü XapI enzimi ile muamele edilmiştir. Sonuç olarak; TGF? geni için sendromsuz dudak damak yarık oluşumu bakımından hasta ve kontrol grubu arasında anlamlı bir fark bulunmuştur (p=.0.0365, p<0.05). Fakat TGFß3 geni için sendromsuz dudak damak yarık oluşumu bakımından hasta ve kontrol grubu arasında anlamlı bir fark bulunamamıştır (p=0.618, p>0.05). Türk toplumunu içeren daha fazla çalışmaya, özellikle her toplumda olduğu gibi Türk toplumunda da dudak damak yarıklı vakaların incelenmesinde çevresel faktörlerin ve bundan sonraki araştırmalarla ortaya çıkarılabilecek moleküler etmenlerin göz önünde bulundurulduğu genetik araştırmalara ihtiyaç vardır. Cleft lip and/or cleft palate is among the most common of all major birth defects, occurring with an incidence of 1/500 to 1/2500 in different populations, varying with geographic location, ethnic group, and socioeconomic conditions. Nonsyndromic cleft lip and/or palate in the formation, most of the studies happened concerning the genetic factors, construct and expression levels to investigate of candidate genes in the animal models. In this study, Turkish population, nonsyndromic cleft lip and palate cases in other population was determined from candidate genes in TGF? ve TGFß3 describe to mutations and to determine this candidate genes frequency in Turkish population. For study, 70 cleft lip and palate cases, 85 control individuals was used. DNA obtained from nonsyndromic cleft lip and palate patients, investigations of TGF? and TGFß3 the PCR product obtained for genes mutations, PCR made of certain exon regions. And RFLP procedures were passed. The PCR product obtained for TGF? gene with HinfI restriction enzyme, the PCR product obtained for TGFß3 with XapI restriction enzyme was treated. As a result, for TGF? gene in terms of the formation of nonsyndromic cleft lip and palate, was found significant difference between patient and control group (p=.0.0365, p<0.05). But for TGFß3 gene in terms of the formation of nonsyndromic cleft lip and palate, was not found significant difference between patient and control group (p=0.618, p>0.05). Turkish society with more work, especially in every society, as well as in Turkish society celft lip and palate cases with analysis of environmental factors and subsequent investigations revealed to be the molecular factors taken into consideration are needed genetic studies.
