Abstract
Olanzapin (OLZ), Risperidon (RPD) ve Ketiapin (QTP) başta şizofreni olmak üzere pek çok ruhsal hastalığın tedavisinde yaygın olarak kullanılan atipik antipsikotik ilaçlardır. Ancak son araştırmalar atipik antipsikotiklerin sinir ve kardiyovasküler sistemler üzerinde toksik etkilere sebep olduğunu ortaya koymaktadır. Bilgilerimize göre, OLZ, RPD ve QTP'nin insan lenfosit kültürlerinde genotoksik hasar potansiyellerine dair çok az bilgi bulunmaktadır. Bu nedenle, mevcut çalışmada OLZ, RPD ve QTP'nin (0 - 400 mg/L) insan tam kan kültürlerinde genotoksik etki potansiyelleri değerlendirildi (n=4). DNA hasarını belirlemek için tek hücre jel elektroforezi (SCGE) ve mikroçekirdek (MÇ) testleri uygulandı. Sonuçlarımız test edilen antipsikotik ilaçların genotoksik etkiye sahip olmadığını ortaya koydu. Nitekim toplam DNA hasarı puanı (SCGE testi) ve MÇ/1000 hücre değerleri ile kontrol grubu değerleri arasında istatistiksel bir fark bulunmadı (p>0.05). Ancak ilaçların yüksek dozları (250 mg/L ve üzeri) kültürlerin üreme koşullarını olumsuz etkiledi. Sonuç olarak atipik antipsikotiklerin kullanımının güvenilir olduğu fakat maruz kalınan dozun artışına bağlı olarak ortaya çıkabilecek sitotoksik etkilerin dikkate alınması gerektiği kanaatine varıldı. Olanzapine (OLZ), Risperidone (RPD) and Quetiapine (QTP) are atypical antipsychotic drugs and are commonly used for the treatment of schizophrenia and other disorders. However, recent reports indicated that these drugs could exhibit toxic effects on nervous and cardiovascular systems. To our best knowledge, there is scarce data considering the genotoxic damage potentials of OLZ, RPD and QTP on human lymphocyte culture system. Therefore, in this study, the genotoxic potentials of OLZ, RPD and QTP (0 to 400 mg/L) have been evaluated in human whole blood cultures (n=4). The single cell gel electhrophoresis (SCGE) and micronucleus (MN) assays were applied to estimate the DNA damage. The results of the present study indicated that the tested antipsychotic drugs did not induce genotoxicity. In fact, the mean values of the total scores of cells showing DNA damage (for SCGE assay) and MN/1000 cell were not found significantly different from the control values (p>0.05). However, the application of the highest drug concentrations (250 mg/L and above) caused to sterility. It is concluded that the tested three different atypical antipsychotic drugs can be used safely, but it is necessary to consider the cyototoxic effects that are likely to appear depending on the increases of doses exposured.
