Abstract
ABSTRACT In this study it has been showed that Zeeman Background Corrected Graphite Furnace Atomic Absorption Spectrometric method, which has been developed for the analysis of selenium in biological materials such as serum, plasma, erythrocyte, whole blood and breast milk, was a reliable, rapid and accurate one which hadn't required any other proprocesses. Since there is a very short range between the essential and toxic levels of selenium which is an essential trace element for organism, the analysis of selenium at ppb level in biological materials is being important. Optimum matrix modifier, pyrolysis and atomization temperatures were determined as Pd+Mg(N03)2, 1200 °C and 2200 °C respectively, in the optimization studies carried out with various matrix modifiers. The optimum Pd and Mg(N03)2 concentrations were found as 300 ppm and 600 ppm respectively. Drying step was achieved by three steps as 70, 120 and 250 °C with temperature ramping programme. Raising to pyrolysis was made by temperature ramping while raising to atomization temperature was achieved at zero second in order to provide the maximum power heating. Cleaning process was made by two step. As a result of these optimization studies the developed furnace programme and the other analysis parameters we used were observed to be in accordance with Stabilized Temperature Platform Furnace conditions. Since the calibration curves of direct calibration and standard addition methods were found in parellel, the direct calibration curve method was prefered in the analysis. Detection limit, linearity and VIhastalıkların teşhisinde, geliştirilen yöntem ile elde edilen nicel sonuçların önemli bir basamak oluşturabileceği görülmektedir. Ayrıca bu hasta gruplarından sağlanacak biyolojik materyallerde, bu yöntem ije^j^yenl^selenyum analizi yapılarak tedavi aşamasında da etkili olabileceği düşünülmektedir.
