Vakfıkebir ekmek hamurundan laktik asit bakterilerinin izolasyonu-identifikasyonu ve ekmek üretiminde kullanılabilme imkanları

Abstract
Araştırmada Vakfıkebir ekmek hamuru örneklerinden toplam 113 laktik asit bakterisi izole ve identifiye edilmiştir. Örneklerde dominant türün Lactobacillus plantarum (%54) olduğu bunu Lactococcus lactis subsp. lactis (%13,2) ve L. brevis (%7,9)' in izlediği belirlenmiştir. Hamurdan izole-identifiye edilen suşlardan seçilen L. plantarum HB75 ve L. brevis EG80, starter kültür olarak kullanılarak 4 grup ekmek (kontrol, L. plantarum HB75, L. brevis EG80 ve L. plantarum HB75 + L. brevis EG80) üretilmiştir. Üretilen ekmeklerin bazı kalitatif özellikleri ve uçucu bileşik profili incelenmiştir. Starter kültür kullanımının ekmek hacmi (p<0,01), kabuk rengi (p<0,05), ağırlığı (p<0,05), ekmek içi pH değeri (p<0,01), yumuşaklık (p<0,01), hidrasyon kapasitesi (p<0,01) ve tekstürel özellikleri (p<0,01 veya p<0,05) üzerine önemli etkiye sahip olduğu belirlenmiştir. Starter kültür kullanımının ekmek hacmini artırdığı ve ekmek içini yumuşattığı tespit edilmiştir. En yüksek ekmek hacmi karışık kültür kullanımında elde edilmiştir. Ayrıca starter kültür kullanımının uçucu bileşikler üzerinde farklı seviyelerde (p<0,01 veya p<0,05) etkili olduğu da saptanmıştır. L. plantarum HB75 + L. brevis EG80 karışık kültürünün hamurda ve ekmek içinde toplam uçucu bileşik miktarı açısından en yüksek değere sahip olduğu görülmüştür. In this research, 113 lactic acid bacteria were isolated and identificated from Vakfıkebir bread dough samples. Dominant species in dough samples was Lactobacillus plantarum (54%) followed by Lactococcus lactis subsp. lactis (13,2%) and L. brevis (7,9%). From the strains isolated and identificated from the dough samples, L. plantarum HB75 and L. brevis EG80 were used as starter cultures and 4 bread groups (control, L. plantarum HB75, L. brevis EG80 and L. plantarum HB75 + L. brevis EG80) were produced. The qualitative properties and volatile compounds profile of breads were investigated. It was determined that the use of starter cultures had a significant effect on volume (p<0,01), crust color (p<0,05), weight (p<0,05), crumb pH (p<0,01), softness (p<0,01), hydration capacity (p<0,01) and texturel properties of breads (p<0,01 or p<0,05). The use of starter cultures increased bread volume and softened bread crumb. The mixed culture (L. plantarum HB75 + L. brevis EG80) bread had the highest volume. It was also determined that the use of starter cultures had a significant effect on volatile compounds in different levels (p<0,01or p<0,05). In terms of total volatile compound amounts, the mixed culture showed highest value in dough and bread crumb.
