Abstract
Bu tezde, Lamiaceae familyasına ait altı farklı türden hidrodistilasyonla elde edilen ve allelopatik potansiyelleri olan esansiyal yağların uygulandıkları mısır tohumları üzerine genotoksik etkileri belirlendi. Elde edilen uçucu yağların içerikleri GC-MS yöntemi ile tayin edildi. Bu yağlar, biyolojik aktivitelerini belirlemek için farklı dozlarda Zea mays L. tohumlarına uygulandı. Bileşenlerin, artan dozlarda mısır tohumlarının çimlenme oranlarıyla beraber kök ucu meristemlerinin mitotik bölünme yüzdelerini düşürdüğü belirlendi. Esansiyel yağların genotoksik etkilerini belirlemek için RAPD ve AFLP teknikleri kullanıldı. Genomik stabilite RAPD, dozlar arasındaki genetik farklılıklar ise AFLP tekniğiyle belirlendi. RAPD ve AFLP profillerindeki değişimlerin çimlenme oranlarıyla mitotik indeks sonuçlarını desteklediği gözlendi. Ancak bu etkinin doza bağlı değişimler sergilemediği tespit edildi. En yüksek polimorfizm oranı C. nepeta bitkisinin uçucu yağlarında 55,44 olarak kaydedilirken, normale en fazla yaklaşan tür O. rotundifolium olup, GTS değeri 85,86 olarak belirlendi. AFLP analizlerinde aynı tür olmalarına rağmen farklı dozlarda uygulanan uçucu yağlardan dolayı mısır örneklerinin genetik benzerlik indeksleri, kontrole göre 0,272 (M. longifolia'da) ile 0,782 (N. nuda'da) değerleri arasında değişim gösterdi. Uçucu yağların gen ekspresyonuna etkisini belirlemek amacıyla da mısır örneklerinin toplam çözünebilir protein miktarları belirlenip SDS-PAGE ile protein profillerinde meydana gelen değişimler belirlendi. Çözünür protein seviyeleri, genotoksisite testlerindeki sonuçlar gibi dozdan bağımsız bir şekilde değişiklik gösterdi. Uçucu yağların benzer şekilde hormon seviyelerini de etkilediği tespit edildi. Genel anlamda esansiyel yağların etkilerini; sitotoksik, genotoksik ve fizyolojik hasarlar oluşturarak gösterdikleri belirlendi. In the present thesis, genotoxic effects of the essential oils obtained by hydro-distillation from six different species of Lamiaceae family were determined against to maize seeds. The obtained volatiles were qualified with GC-MS. To determine their biological activity, the oils were applied to Zea mays L.seeds at different doses. It was observed that germination rates and mitotic division of the root tips were decreased with the increasing of the applied compounds? doses. The genotoxicity of the volatiles was determined by using RAPD and AFLP techniques. Genomic stability was estabilished by RAPD and the genetic differences between doses with AFLP technique. The alterations obtained from RAPD and AFLP profiles supported the germination rate and mitotic index results. While the highest polymorphism value was calculated as 55,44 for C. nepeta volatiles, the closest species to control was O. rotundifolium with 85,86 GTS value. Due to different applied concentrations of volatile oils, the genetic similarity indices of maize seedlings varied from 0,272 (M. longifolia) to 0,782 (N. nuda), even though they were same species according to the AFLP analyses. In order to determine effects of the volatiles on gene expression, the soluble protein levels of the samples were measured and the changes occured in the protein profiles were determined by SDS-PAGE analyses. Similar to the genotoxicity results, the soluble protein levels of the samples showed differences without a dose-dependent manner. Samely, hormone?s level was affected by the volatiles. In general, these compounds showed their effects as a result of their cytotoxic, genotoxic and physiological damages.
