Yenilebilir bazı böceklerin ın vitro genotoksik ve oksidatif etkilerinin incelenmesi

Abstract
İnsanoğlu yüzyıllardan beri böcekleri önemli bir besin kaynağı olarak tüketmiştir. Modern çağlarda ise böcek yeme birçok toplulukta azalmıştır. Ancak dünya çapında dağılmış olan çeşitli kültürler arasında böcekler protein, yağ, mineral ve vitaminlerin ana kaynağı olan ve tercih edilen bir gıda halindedir. Yenilebilen böceklerin dünyada bu kadar yaygın olmalarına karşın literatürde genotoksik ve oksidatif etkileriyle ilgili sınırlı bilgi mevcuttur. Bu çalışmada, Güneybatı Nijerya'da yenen iki yaygın böcek türü olan Zonocerous variegatus (Orthoptera: Pyrgomorphidae) ve Oryctes boas (Solanales Solanaceae)'ın sulu özlerinin sitogenetik ve oksidatif etkileri insan tam kan hücre kültürleri üzerinde değerlendirildi. Özler, çeşitli konsantrasyonlarda (0 ppm'den 2000 ppm'e kadar) kültürlere ilave edildi. DNA ve kromozom hasarı potansiyelleri bulmak için Kromozom aberasyon (KA) ve Mikroçekirdek (MÇ) testi kullanıldı. Oksidatif etkilerini değerlendirmek amacıyla total antioksidan kapasite (TAK) ve toplam oksidan durum (TOD) düzeyleri ölçüldü. Sonuçlarımız test edilen konsantrasyonlarda genotoksik etkilerinin olmadığını gösterdi. Ancak, özler TAK ve TOD seviyelerinde doza bağlı değişikliklere neden oldu. Mevcut bulgulara dayanılarak, incelenen böceklerin güvenle tüketilebileceği sonucuna varıldı. Ancak oksidatif strese bağlı olarak görülmesi muhtemel hücresel zararların dikkate alınması gerekmektedir. Ayrıca yenilebilir böceklerin potansiyel sağlık risklerinin değerlendirilmesinde oksidatif ve genotoksik analizleri içeren bu in vitro yaklaşımın faydalı olabileceği düşünülmektedir. Humans have consumed insects as an important food resource for centuries. In modern times, eating insects has declined in many societies. Yet, among various cultures scattered throughout the world, insects remain a preferred food and an essential source of protein, fat, minerals and vitamins.Edible insects have become so widespread in the world, although very limited information is available concerning with their genotoxic and oxidative effects in the literature. In this study, the cytogenetic and oxidative effects of water soluble extracts of two commonly eaten insects in Southwestern Nigeria; Zonocerous variegatus (Orthoptera: Pyrgomorphidae) and Oryctes boas (Solanales: Solanaceae) were evaluated on whole cultured human blood cells.The extracts were added into the cultures at various concentrations (0 to 2000 ppm).The chromosome aberration (CA) and micronucleus (MN) tests were used tofind out the DNA and chromosomal damage potentials by aqueous insect extracts in vitro. To assess the oxidative effects total antioxidant capacity (TAC) and total oxidant status (TOS) levels were also measured. Our results indicated that these extracts did not show genotoxic effects at all tested concentrations. However, the extracts caused dose dependent alterations in both TAC and TOS levels. Based on the findings, it was concluded that the studied insects can be consumed safely, but it is necessary to consider the cellular damages which are likely to appear depending on the oxidative stress. We also suggest that this in vitro approach for oxidative and genotoxicity assessments may be useful to compare the potential health risks of edible insects.
