Abstract
Çalışmamızda, in vitro koşullar altında kuraklık stresinin buğday (Triticum aestivum) genomunda yaptığı genetiksel ve epigenetiksel değişimler üzerine selenyumun etkileri araştırılmıştır. Tohumlardan elde edilen olgun embriyolar, PEG 6000 ile oluşturulan 4 farklı osmotik potansiyelde (0, %5, %10, %15) ve Na2SeO4'ün 4 farklı konsantrasyonunu içeren (0, 2, 4 ve 6 µM) MS ortamında kültüre alınmıştır. Deney gruplarının RAPD profillerindeki DNA değişimleri, DNA hasarları ve mutasyonlar ile ilgili bilgi verirken CRED-RA yöntemi DNA metilasyonunu hakkında bilgi vermiştir. Sonuç olarak, kuraklık stresinin DNA hasarlarına ve GTS (Genomic template stability) değerinin düşmesine neden olduğu ve DNA hipermetilasyonu oluşturduğu gözlenmiştir. Kuraklık ile beraber uygulanan selenyumun, DNA hipometilasyonuna yol açtığı ve düşük dozlarının (2, 4 µM Na2SeO4) kuraklık sonucu oluşan GTS değerlerini yükselttiği, yüksek dozunun ise (6 µM Na2SeO4) GTS değerlerini azalttığı gözlenmiştir. In our study, the effect of selenium on genetic (RAPD) and epigenetic changes (CRED-RA) in wheat (Triticum aestivum) exposed to drought stress in vitro conditions were investigated. The embryos obtained from seeds were cultured on medium (MS) consisting of 0, 5%, 10%, 15% PEG 6000 and 0, 2, 4 ve 6 µM Na2SeO4. DNA changes of RAPD (Randomly amplified polymorphic DNA) profiles in the experimental group exhibited DNA damage and mutations (eg, point mutations and chromosomal arrangements) and CRED-RA method was performed to determine the changes of DNA methylation. In conclusion, drought stress caused DNA damage by reducing in the values of the GTS (Genomic template stability) and DNA hipermethylation. When selenyum was applied against drought stress, selenyum ameliorated the effect of drought. The low doses of selenium (2, 4 µM Na2SeO4) increased reduction of GTS values caused by drought, whereas the high dose of selenium (6 µM Na2SeO4) decrease in the GTS of wheat genome exposed to drought
