Abstract
Bu tezde, Ağrı/Diyadin kaplıcasından izole edilen, konvensiyonel ve moleküler yöntemler kullanılarak karakterize edilen Brevibacillus sp. (Z1) (Gen Bank No:KC292196.1) bakterisinden ekstraselüler lakkaz enzimi sırasıyla; amonyum sülfat çöktürmesi, DEAE sefadeks iyon değişim kromatografisi ve sephadeks G100 jel filtrasyon kromatografisi basamakları kullanılarak saflaştırıldı. Saflaştırılan enzimin optimum pH, optimum sıcaklık, Vmax-Km değerleri, ayrıca sıcaklık ve pH stabiliteleri belirlendi. Enzimin ABTS, 2,6-dimetoksifenol ve guaikol substratlarına karşı substrat spesifikliği belirlendi. Enzim üzerine Ca2+, Mg2+, Hg2+, Mn2+, Na+, Zn2+, Cu2+, Cd2+, Co2+ ve Fe2+ gibi bazı metal iyonlarının ve EDTA, sodyum azid, dithioeritritol ve SDS gibi bazı kimyasalların etkisi araştırıldı. Saflaştırılan lakkaz enziminin endüstriyel süreçlerde ve de çevresel atıkların biyolojik olarak yıkımında kullanılabilirliğinin araştırılması amacıyla ticari olarak satılan ve tekstilde kullanılan bazı boyaların enzimatik olarak dekolorizasyonu çalışıldı.Gerçekleştirilen deney sonucunda elde edilen verilere göre; lakkaz enziminin optimum pH'sı 4.0, optimum sıcaklığını 60oC, molekül ağırlığı 122.02, 93.756 ve 73.790 kDa büyüklüğünde, 3 alt birimden oluştuğu tespit edildi. ABTS ve 2,6-dimetoksifenol substratları için Vmax değerleri sırasıyla 14.08 EU/mL.dak., 16.12 EU/mL.dak., Km değerleri ise ABTS, 2,6-dimetoksifenol substratları için sırasıyla 5.34 mM ve 3.03 mM olarak hesaplandı. Brevibacillus sp. (Z1)'den ekstraselüler olarak üretilip saflaştırılan lakkaz enzimini kullanarak çevre kirliliğine sebep olan Reaktif Siyah 5, Fuksin, Allura Kırmızısı ve Asit Kırmızısı 37 gibi bazı tekstil boyalarının giderilmesinde kullanılabilirliği araştırıldı. Enzimin Reaktif Siyah 5, Fuksin, Allura Kırmızısı ve Asit Kırmızısı 37 boyalarının 3 saat sonunda %66.0, %50.8, %53.6 ve %47.3 oranında azalttığı belirlendi.Anahtar Kelimeler: Lakkaz, Moleküler karakterizasyon, Brevibacllus sp., Dekolorizasyon, Saflaştırma, Karakterizasyon In this thesis, thermophilic bacterial isolates were obtained from Ağrı/Diyadin hot spring. Then, the laccase activities of these isolates were inestigated to find the isolate, which has has the highest laccase activity. The isolate with the highest laccase activity was identified as Brevibacillus sp. (GEN BANK NO: KC292196.1) by using conventional and molecular methods. The extracellular laccase enzyme of Brevibacillus sp. was purified by using respectively ammonium sulfate precipitation, DEAE cellulose ion exchange chromatography and Sephadex G100 gel filtration chromatography steps. Optimum pH, optimum temperature, Vmax-Km values and separately temperature and pH stabilities of the purified enzyme were determined. The substrate specificity of the enzyme was determined against to the substrates of ABTS, 2,6-dimethoxyphenol and guaicols. The influence of some metal ions Ca2+, Mg2+, Hg2+, Mn2+, Na+, Zn2+, Cu2+, Cd2+, Co2+ and Fe2+ some chemicals such as EDTA, sodium azide, dithioeritritol, SDS on the enzyme was studied. With the purpose of the investigation of the purified laccase enzyme in industrial processes and also in the biological breakdown of environmental waste, decolourising enzymatically of some dyes which are available in textile and commercially was studied. According to the data obtained as a result of the performed experiments, it has been detected that the optimum pH of laccase enzyme is 4.0, its optimum temperature is 60'C and its molecular weight is 122.02, 93.756 ve 73.790 kDa. It has been calculated that V-max values for ABTS and 2,6-dimethoxyphenol substrates are respectively 14.08 EU/mL.dak., 16.12 EU/mL.dak. ; Km values for those substrates are respectively 5.34 mM, 3.03 mM. It was researched whether the purified enzyme which has been produced as extracellular may be used for the removing of some textile dyes such as Reactive Black 5, fuchsin, Allura red and Acid Red 37 which has caused environment contamination or not. It was determined that the enzyme reduces Reactive Black 5 enzyme, Fuchsin, Allura Red and Acid Red 37 dye to %66.0, %50.8, %53.6 and %47.3 at the end of 3 hours.Keywords: Laccase, Molecular Characterization, Brevibacillus sp., Decolorization, Purification, Characterization
