Abstract
Bu çalışmada Ayder Kaplıcaları'ndan izole edilen Bacillus licheniformis A10'dan ekstraselüler alkalin proteaz enzimi saflaştırıldı ve karakterize edildi. Enzim saflaştırılmasında amonyum sülfat çöktürmesi-diyaliz ve DE52 anyon değişim kromotografisi yapıldı. Proteaz enzimi %9,44 verimle 1,38 kat saflaştırıldı. Enzimin SDS-PAGE ile molekül ağırlığı yaklaşık 40,55 kDa olarak tespit edildi. Enzim için optimum pH ve sıcaklık değerleri 9 ve 70ºC olarak belirlendi. Bacillus licheniformis'e ait saflaştırılan proteaz enzim aktivitesi üzerine metallerin etkisi incelendiğinde; Mg+2, Mn+2, K+ varlığında enzimin aktivitesinin arttığı gözlenirken; Fe+2, Zn+2'nin aktiviteyi azalttığı, Ca+2'nin ise aktivitede değişime neden olmadığı görüldü. Enzimin oksidantlar, yüzey aktif maddeleri ve organik çözücüler varlığında aktivite kaybettiği tespit edildi. Substrat spesifikliği çalışmalarında; kazein, azokazein, jelatin ve bovin serum albumin (BSA) arasında enzimin en yüksek aktiviteyi kazein için gösterdiği gözlendi. Enzimin serin proteaz inhibitörü fenilmetilsülfonilflorid (PMSF) ile tamamen inhibe olduğu belirlendi. Lineweaver-Burk grafiğinden yararlanılarak tespit edilen KM ve Vmax değerleri 0,033 mg/ml ve 8,17 µmol.ml-1.dk-1 olarak elde edildi. In this thesis, purification and characterization of a protease produced by Bacillus licheniformis A10 isolated from Ayder Hot Spring. Ammonium sulfate precipitation, dialysis and DE52 anion-exchange chromatography were performed to purify the enzyme. Protease enzyme was purified 1,38 fold with 9,44% yield. Molecular mass of the enzyme showed approximately 40,55 kDa with SDS-PAGE. Optimal pH and temperature were determined for the enzyme as 9,0 and 70ºC, respectively. The effects of various metal ions (Mg2+, Mn2+, K+, Fe2+, Zn2+, Ca2+) on the protease enzyme activity were examined. According to our results, while the enzyme activity increased in the presence of Mg2+, Mn2+, K+, the enzyme activity decreased in the presence of Fe2+, Zn2+. Howevere, no changes were seen in the presence of Ca2+. It is also observed that the enzyme loses its activity in the presence of surfactants, oxidants and organic solvents, but show highest activity towards casein relative to other native proteins such as gelatin, ceratin, egg albumin and BSA. The enzyme was completely inhibited by PMSF which is a general serine protease inhibitör. The kinetic parameters, Vmax and KM, of the purified protease were determined by using Lineweaver-Burk plot 0,033 mg/ml and 8,17 µmol. ml-1.dk-1 respectively.
