Abstract
Kolorektal kanser dünya genelinde kanserle ilişkili ölümlerin önemli bir nedenidir ve en sık görülen üçüncü kanser türüdür. Kolorektal kanserin klinik tedavisinde 5-Flourouracil (5-FU) temelli uygulamalar ana basamağı oluşturmaktadır. Ancak 5-FU'nun antitümör etkisinin yanı sıra yüksek toksisite ve yan etkilere sahip olması, 5-FU temelli yeni tedavilerin geliştirilmesini gerekli kılmaktadır.Bu çalışmada, HT-29 kolorektal adenokarsinoma hücre hattı üzerinde iki farklı bitkisel etken maddenin (kuersetin ve luteolin) ve iki bitki türünden (Pistacia eurycarpa ve Achillea biebersteinii) polaritesi farklı çözücüler kullanılarak hazırlanmış bitki ekstrelerinin 5-Flourouracil ile kombinasyonlarının sinerjistik etkileri araştırıldı.MTT testi ile hücrelere ait % canlılık oranları belirlendi. Uygulanan tedavi gruplarına ait IC50 değerleri hesaplandı. IC50 değerlerine göre bitkisel etken madde ve ekstreler tek başlarına ve 5-FU ile kombine olarak hücrelere uygulandı. Bu kombinasyonların etkinlikleri izobologram analizi ile saptandı. Hücrelerin apoptoza yönelme oranları CDDE yöntemi ve apoptotik indekslerin hesaplanması ile belirlendi. VEGF miktarlarındaki azalma oranları Hu VEGF ELISA yöntemi ile hesaplandı. Uygulanan tedavilerin, apoptotik yolaklarda önemli rollere sahip olan p53, Bcl-2, Bax, mTOR, Akt, PTEN, p38 MAPK gen ve proteinleri üzerindeki etkileri RT-PCR ve western blot yöntemi ile analiz edildi. HT-29 kolorektal adenokarsinoma hücrelerinin canlılığının azalması ve apoptoza yönelmesinde, 5-FU ile bitkisel etken madde ve ekstrelerin kombinasyon olarak uygulanması, 5-FU'nun tek başına uygulanmasına göre daha etkili oldu. Sonuç olarak, uygulanan kombine tedavilerde 5-FU'nun düşük dozlarda bile etkinliğinin arttığı ve böylece ilacın yüksek sitotoksik etkilerinin minimuma indirilebileceği saptandı. Colorectal cancer is a leading cause of death world-wide associated with cancer and is the third most common type of cancer. In the clinical treatment of colorectal cancer, 5-Fluorouracil (5-FU) based applications are the main steps. However, because 5-FU has anti-tumor effect as well as high toxicity and adverse effects, the development of new therapies based on 5-FU becomes necessary.In this study, synergistic effects of the combination with 5-Flourouracil of two different herbal active substances (quercetin and luteolin) and two plant species (Pistacia eurycarpa and Achillea biebersteinii) of plant extracts whose polarity was prepared using different solvents over HT-29 colorectal adenocarcinoma cell line were investigated.Percentage survival rates of cells were determined by MTT test. IC50 values of the treatment groups were calculated. According to the IC50 value, plant extracts and herbal active ingredients were applied to cells alone and then in combination with 5-FU. Efficacy of this combination was determined by isobologram analysis. Ratio orientation to apoptosis of cells was determined by CDDE method and calculating the apoptotic index. Proportional reduction in the amount of VEGF was calculated using Hu VEGF ELISA. The effects of the applied treatment onp53, Bcl-2, Bax, mTOR, Akt, PTEN, p38 MAPK genes and proteins which have an important role in the apoptotic pathway were analyzed by RT-PCR and western blot method.In the reduction of the viability of HT-29 colorectal adenocarcinoma cells and in orientation toward apoptosis, application of the 5-FU in combination with herbal active ingredients and extracts was more effective than 5-FU therapy alone. As a result, in combined treatments, it was reached out that the efficiency of 5-FU increases even at low doses and that high cytotoxic effect of the drug can be minimized.
