Abstract
Araştırmada 20 farklı firmadan temin edilen pastırma örneklerinden toplam 271 enterokok izolatı bazı fenotipik özellikler yönünden (farklı sıcaklıklarda gelişme, glukozdan gaz üretimi, farklı pH değerlerinde gelişme, pigmentasyon, piruvat aktivitesi, hareket, hemoliz) test edilmiş ve seçilen 100 izolat fenotipik (VITEK 2GP) ve genotipik (16S rRNA) olarak identifiye edilmiştir. Bu 100 suş antagonistik aktivite yönünden de test edilmiştir. Ayrıca pastırma örnekleri mikrobiyolojik (laktik asit bakterileri, Enterococcus, Micrococcus/Staphylococcus, maya-küf, Enterobacteriaceae) sayımlar ile pH ve nem analizlerine de tabi tutulmuştur. Örneklerde dominant florayı genellikle Micrococcus/Staphylococcus oluşturmuştur. Laktik asit bakteri ve maya-küf sayıları sırasıyla 103-108 kob/g, 102-106 kob/g arasında değişmiştir. Enterobacteriaceae ise dört örnek hariç saptanabilir sınırın altında tespit edilmiştir. Enterokok sayısı ise örneklerin %35'inde 1x102-<1x104 kob/g aralığında yer alırken diğer %35'lik kısım ise 106 kob/g'dan daha yüksek sayılar vermiştir. Pastırma örneklerinde dominant türün E. faecium olduğu belirlenmiştir. İdentifiye edilen 100 izolattan sadece 9'u kuyu difüzyon testinde L.monocytogenes'e karşı antagonistik aktivite göstermiştir. Suşlar S.aureus ve E.coli O157:H7'ye karşı ise antagonistik aktivite sergilememiştir.Anahtar Kelimeler: Pastırma, Enterokok, İdentifikasyon, Starter Kültür, Antagonistik Aktivite A total of 271 Enterococcus isolates obtained from pastırma samples from 20 different firms were tested for some phenotypic properties (growth at different temperatures, gas production from glucose, growth under different pH values, pigmentation, pyruvate activity and hemolysis) and 100 selected isolates were identified as phenotypic (VITEK 2GP) and genotypic (16S rRNA). These 100 strains were also tested for antagonistic activity. In addition, pastırma samples were also subjected to microbiological (lactic acid bacteria, Enterococcus, Micrococcus/Staphylococcus, yeast-mold, Enterobacteriaceae) counts and pH and moisture analyses. In general, dominant flora in the samples was constituted by Micrococcus/Staphylococcus. Lactic acid bacteria and yeast-mold counts were varied between 103-108 kob/g and 102-106 kob/g, respectively. Except for four samples, Enterobacteriaceae were determined to be under the detectable level. Enterococcus counts were found to be in 1x102-<1x104 kob/g in range 35% of the samples while another 35% were counted to be greater than 106 kob/g. It was determined that the dominant species in pastırma samples was E. faecium. Only 9 of the 100 identified isolates showed antagonistic activity against L.monocytogenes in well diffusion test. No antagonistic activity against S.aureus and E.coli O157:H7 were observed.Keywords: Pastırma, Enterococcus, Identification, Starter Culture, Antagonistic Activity
