Abstract
IV ÖZET Bu çalışmada 4-nitroquinolin-N-oxid (4NQ0) ve mitomisin C (MMC)'nin mutajenik potansiyellerine karşı riboflavin ve redükte glutatyonun antimutajenik etkilerinde antioksidant enzimlerin rolü incelenmiştir. Bu amaçla riboflavin ve glutatyon uygulanmış bakterilerde süperoksit dismutaz (SOD) ve katalaz aktiviteleri ve aynı zamanda Salmonella/mikrozom (Ames) test sistemi kullanılarak aynı maddelerin, mutajenite üzerine etkileri saptanmış ve sonuçlar karşılaştırmıştır. Mutajenik etkinin antimutajen maddelerce önlenmesi sırasında Salmonella typhimurium TA 100 ve TA 102 mutant suşlarında SOD ve katalaz enzim düzeylerinin saptanmasıyla, antioksidant enzim düzeyleri ve mutajenite arasında bir bağlantı olup olmadığı araştırılmıştır. Riboflavin, Salmonella typhimurium TA 100 ve TA 102 suşlarında 4NQO'in mutajenitesine antimutajenik etki gösterirken (inhibisyon %40-60 arasında), MMC mutajenitesine etki etmemiştir. Glutatyon ise, her iki susta da 4NQO ve MMC mutajenitesini artırmıştır. Riboflavinin antimutajenik etkisinde antioksidant enzimlerden SOD ve katalaz aktivitesinde değişiklik saptanmamıştır. Aynı durum glutatyon için de geçerlidir. Çünkü her iki durumda da katalaz ve SOD aktivitelerinde 4NQO ve MMC mutajenitesine bağlı olarak bir değişim gözlenmediği gibi antimutajenik etkiye bağlı bir değişim de saptanmamıştır. ABSTRACT In this study the role of antioxidant enzymes on the antimutagenic actions of riboflavin and glutathione against mutagenic potentials of 4NQO and MMC have been investigated. For this purpose the activities of superoxide dismutase (SOD) and catalase have been detected in bacterial cultures treated with riboflavin and glutathione. By Ames Salmonella/microsome test system the effect of those antimutagens on mutagenic actions of 4NQO and MMC have been detected. In Salmonella typhimuriumTM 02 and TA 100 mutant strains the activities of SOD and catalase during the prevention of mutagenic action by antimutagens have been detected. The correlation between the antioxidant enzyme levels and mutagenecity have been investigated. While riboflavin displayed antimutagenic effect (% 40-60 inhibition) on 4NQO mutagenicity in Salmonella typhimurium TA1 02 and TA1 00 strains, it did not have any effect on the mutagenicity of MMC. Glutathione increased 4NQ0 and MMC mutagenicity in both strains. The antioxidant enzymes SOD and catalase seemed to have no effect on the antimutagenic action of riboflavin. The same is true for glutathione, for in both conditions no change in the activities of catalase and SOD in connection to 4NQ0 and MMC mutagenicity were detected.
