Abstract
IV ÖZET Sunulan tez çalışması kapsamında grubumuzda daha önce üretilmiş olan STM'in (Scanning Tunnelling Microscopy) geliştirilerek kullanım kolaylığı ve kaitesi artırılmış ve geliştirilen sistemin performansı araştırılmıştır. Bu amaçla birinci bölümde STM sisteminin donanım ve yazılım bölümlerinde yeni tasarım ve imalatlar yapılmıştır. Donanım bölümünde: (i) Tarama alanını ve görüntü kalitesini artırmak amacıyla ± 250 V (500 V) bilgisayar kontrollü bipolar güç kaynağı tasarlanıp imal edilerek elektronik açıdan performansı araştırılmıştır, (ii) Bağıl olarak iletkenliği kötü olan biyolojik molekülleri görüntüleyebilmek, sıvı ortamda çalışabilmek amacıyla akım/gerilim çeviricisinin ölçüm alanı (pA-nA) genişletilmiştir, (iii) Tarama hareketinin doğrusallığının artırılması amacıyla, tarama iğnesi piezo tarayıcının ortasına alınmış, bu amaçla bir vida mekanizması imal edilmiştir, (iv) Sıvı örneklerle çalışabilmek amacıyla düşey yönde hareketli bir örnek tutucu tasarlanıp imal edilmiştir, (v) Görüntü işleme olanaklarını artırmak amacıyla çoklu işletim ortamında çalışabilecek yeni bir kontrol programı yazılmıştır. İkinci bölümde ise tüm sistemin performansını araştırmak amacıyla yapay ve doğal polimerik moleküllerden örnekler seçilerek bunların STM görüntüleri alınmıştır. İpliksi yapıda olan doğal moleküllerden kolajen, globular yapıda olanlardan albumin, antibadiler (IgG ve IgM) ve amino asitlerin tamamı (20 adet) görüntülenmiştir. STM çalışmalarında DNA, model olarak en çok kullanılan biyomoleküldür. Sunulan tez kapsamında ise DNA'mn çeşitli boya ve ilaçlarla etkileşimi görüntülenmiştir. Sentetik polimerlerden ise P(MMA/BMA) nanopartiküller ve bunların film oluşturma özellikleri STM görüntüleri ile araştırılmıştır. Yine sentetik polimerlerden P(NIPAAm-Aac) uyan cevap polimerleri ve amino asitlerin bu polimerlere bağlanarak oluşturdukları yapılar STM ile görüntülenmiştir. Bütün deneyler sırasında optimum görüntüleme parametreleri, substrat, iğne türü, çalışma akımı, iğne-örnek öngerilimi açısından araştırılarak sunulan tez içinde rapor edilmiştir. ABSTRACT In the content of the proposed thesis; the STM (Scanning Electron Microscopy) that was constructed before by our group was improved, its quality and using advantages were increased, and the performance of the developed system was tested. In the first part of the thesis, software and hardware of the STM were improved. In the hardware: (i) To increase the scanning area and imaging quality, ± 250 V (500 V) bipolar, computer controlled power supply is designed and constructed. Then the performance of the power supply was tested, (ii) For the imaging of the biological molecules mostly has poor conductivity, the measurement range (pA-nA) of the I/V converter was increased, (iii) The tip was installed at the middle of the piezo scanner to improve the lineerity of the scanner. By that purpose, a screw mechanism has been constructed, (iv) To study with liquid samples, a sample holder which has movement in horizontal dimension has been designed and constructed, (v) A new software that works in multi-tasking operating system has been developed to improve the imaging process. In the secand part of the thesis, the images of the chosen natural and synthetic polymeric materials were taken for testing the performance of the whole system. The images of selected natural polymers (the cologen in fibrous form and albumine in globular form) were taken. In other part of the study, the images of the all amino acids which are found in human body were taken. Although the DNA is the most studied biological molecule in the SPM technics, the interaction of the DNA with dies and drugs were studied in the scope of this thesis. Synthetic polymers in nanoparticule form like P(MMA/BMA) and their film formation properties were observed by STM. In similar way, the P(NIPAAm-Aac) from stimuli-responsive polymers and its congugates with amino acids were also observed by STM. The optimum imaging parameters of STM, the type of the tip, substrates, tip-sample bias and operating current were optimised and reported in the scope of the thesis.
