Abstract
ELMA SUYU KONSANTRELERİNDE PATULİN, FUMARİK ASİT VE LAKTİK ASİT OLUŞUMU ÜZERİNE ARAŞTIRMALAR Esma Elden Taydaş Hacettepe üniversitesi, Gıda Mühendisliği Anabilim Dalı ÖZ Bu çalışmada, `Golden Delicious` çeşidi, sağlam elmalar hammadde olarak kullanılmış, Rhizopus stolonifer, Penicillium expansum ve Lactobacillus plantarum saf kültürleri ile aşılanmış ve laboratuvar koşularında elma suyu konsantresi (ESK) üretilmiştir. Elde olunan ESK örneklerinde patulin, laktik asit ve fumarik asit miktarı üzerine mikroorganizmaların ve çeşitli proses uygulamalarının etkileri araştırılmıştır. R. stolonifer, fumarik asit ve laktik asit oluşumuna neden olan etken olarak bulunmuştur. L plantarum'un da laktik asit ürettiği ve malolaktik fermentasyon sırasında karmaşık bir biyokimyasal yol ile az miktarda fumarik asit oluşturduğu belirlenmiştir. P. expansum'un ise sadece patulin ürettiği, örneklerin laktik asit ve fumarik asit içeriğini etkilemediği tesbit edilmiştir. Çatışmada, endüstriyel ESK üretimi sırasında, elma suyunda bulunan patuline farklı proseslerin etkisi de araştırılmıştır. Elma suyundan patulinin uzaklaştırılmasında, döner vakum filtre kullanılarak yapılan geleneksel durultma (berraklaştırma), UF (ultrafiltrasyon) işleminden çok daha etkili olarak bulunmuştur. Geleneksel yöntemle endüstriyel ESK üretimi sırasında elma suyu içinde bulunan fumarik aside işleme teknolojisinin etkisi de araştırılmıştır. Depektinizasyon ve evaporasyon aşamalarında örneklerde, fumarik asit miktarında bir artış belirlenmiştir. Çalışmada aylara göre (Eylül, Ekim, Kasım ve Aralık) ESK örneklerinde, patulin, fumarik asit ve DL-laktik asit düzeyleri incelenmiş ve Eylül ayında bu değişkenlerde önemli bir artış belirlenmezken, Ekim ayında ciddi bir artış gözlenmiş, Kasım ayında diğer aylara göre artışlar maksimum değerlere ulaşmış ve Aralık ayında ise Kasım ayına göre bir düşüş saptanmıştır. The effects of these microorganisms and various processing treatments on patulin, lactic acid and fumaric acid contents of AJC were investigated. R. stolonifer was found to be the most important causative agent as the fumaric acid and lactic acid producer. L. plantarum also produced lactic acid and low amounts of fumaric acid during malolactic fermentation by complicated biochemical pathway. P. expansum produced only patulin without any noticeable effect on lactic acid and fumaric acid contents of apple juice. In this study, the effects of different processing technologies on the patulin and fumaric acid contents of apple juice were also studied in industrial scale. The conventional clarification using rotary vacuum filter was found more efficient for the removal of patulin than the ultrafiltration process. The effect of processing technology on fumaric acid content of AJC was studied through conventional process. The fumaric acid levels of apple juices increased during depectinization and evaporation stages through conventional process. The change of patulin, fumaric acid and DL-lactic acid levels of commercial AJC were also studied through a processing season starting at September and ending at December. In these variables, while insignificant increase were detected on September, obvious increase were detected on October. Maximum values were obtained on November. The values obtained on December were started to decrease when compared to values of November.
